摘    要:目的 研究眼镜蛇毒因子(CVF)激活补体系统,对兔脑出血后脑水肿及肿瘤坏死因子(TNF-α)、神经元特异性烯醇化酶(NSE)的影响.方法 56只日本大耳兔随机分为4组,实验组兔按0.5mg/kg腹腔内注射含CVF的0.9%氯化钠溶液约5ml,对照组兔腹腔注射5m10.9%氯化钠溶液.再设空白组、标准组.4组不同时间点检测血浆补体C3、TNF-α、NSE浓度,以脑(湿-干)/湿检测脑水的含量.结果 兔血浆中补体C3、NSE、TNF浓度,在4组间、组内各时点间比较差异有统计学意义(P〈0.05).左大脑(湿-干)/湿比重:实验组(80.19±0.54)%,对照组(79.57±0.44)%,差异有统计学意义(P〈0.05).结论 CVF能有效激活并耗竭补体,补体系统过度激活加重脑水肿的发生.脑出血急性期TNF-α、NSE升高的峰值与脑损伤、脑水肿严重程度可能存在某种联系,TNF-α、NSE水平可在一定程度上可反应脑损伤和脑水肿情况.

Abstract:Objective To investigate the effects of cobra venom factor (CVF) on hemorrhagic cerebral edema in rabbits. Methods Fifty six Japanese white rabbits were randomly divided into 4 groups (experimental group, control group, blank group and standard group). Cerebral hemorrhage was induced in rabbits of experimental and control groups. Rabbits in experimental group were given intraperitoneal injections of CVF; while those in control group were given intraperitoneal injections of normal saline. The dry weight/wet weight of brain and plasma concentrations of complement C3, neuron-specific enolase (NSE), and tumor necrosis factor (TNF-α ) were measured in all groups. Results There were significant differences in plasma complement C3, NSE and TNF concentration among 4 groups at each time points (P〈0.05). The dry weight/wet weight of brain of experimental group and control group was (80.19±0.54) %and (79.57 ± 0.44)% (P〈0.05). Conclusion CVF can activate complement system and aggravate brain edema after cerebral hemorrhage, which is related to plasma concentrations of TNF- α and NSE.
