柴胡理中汤改善非酒精性脂肪肝病大鼠肝功能和脂代谢效果观察

目的:探讨柴胡理中汤对非酒精性脂肪肝病(Non-alcoholic Fatty Liver Disease,NAFLD)大鼠肝功能和脂代谢的效果。方法:将40只SD大鼠随机分为4组:空白对照组、模型对照组、西药对照组、柴胡理中汤组,每组10只。除空白组外,其余大鼠均饲以高脂饲料8周。8周后,每日以固定时间给予为期12周的相应药物灌胃(Irrigation,Ig),空白组与模型组蒸馏水灌胃,西药组予550 mg·kg-1·d-1多烯磷脂酰胆碱灌胃,柴胡理中汤组予9 g·kg-1·d-1的柴胡理中汤灌胃。实验过程中观察比较各组大鼠一般情况;实验结束前禁食12 h,称取体质量,并计算肝指数;抽取空腹血分离血清后测定肝酶系、血脂;在光镜下观察肝细胞并对其脂肪变性程度评级。结果:西药组与柴胡理中汤组的大鼠一般情况优于模型组,两组的体质量、肝指数、肝酶水平丙氨酸转氨酶(Alanine Aminotransferase,ALT)、门冬氨酸转氨酶(Aspartate Aminotransferase,AST)、谷氨酰转肽酶(Glutamyl Transpeptidase,GGT)]、血脂总胆固醇(Total Cholesterol,TC)、甘油三酯(Triglyceride,TG)、低密度脂蛋白胆固醇(Low Density Lipoprotein Cholesterol,LDL-C)]也明显低于模型组,差异有统计学意义(P<0.05),且两组均能改善脂肪变性,以柴胡理中汤组改善效果更佳。结论:柴胡理中汤能有效降低NAFLD大鼠肝酶水平,改善脂代谢紊乱和肝组织病变,达到治疗大鼠NAFLD的效果。

Efficacy of the Chiahu Lizhong decoction on liver function and lipid metabolism in rats with NAFLD

Objective:To investigate the effect of the Chaihu Lizhong decoction(柴胡理中汤)on liver function and lipid metabolism in rats with nonalcoholic fatty liver disease.Methods:40 SD rats were randomly divided into the blank group,model control group,western medicine control group and the Chaihu Lizhong decoction group,10 rats in each group.Except for the blank group,the other rats were fed a high-fat diet for 8 weeks.After 8 weeks,the corresponding drug was administered at a fixed time for 12 weeks by irrigation,the blank group and the model group were given distilled water,the western medicine group was given 550 mg·kg-1·d-1 polyene phosphatidylcholine,the Chaihu Lizhong decoction group was given 9 g·kg-1·d-1 the Chaihu Lizhong decoction.Results:Conditions in the western medicine group and the Chaihu Lizhong decoction group were generally better than the model group.The body weight,liver index,liver enzyme level(ALT,AST,GGT)and blood lipids(TC,TG,LDL-C)were lower in the western medicine group and the Chaihu Lizhong decoction group than the model group(P<0.05),and both groups can improve steatosis,and the improvement effect was best in the Chaihu Lizhong decoction group.Conclusion:The Chaihu Lizhong decoction can effectively reduce the liver enzyme level of NAFLD rats,improve lipid metabolism disorder and liver tissue lesions.
