芎附散作用机制的网络药理学分析

摘    要:目的基于网络药理学方法分析芎附散活性成分、靶点、通路之间的关系,探讨其作用机制。方法依托BATMAN-TCM数据库和TCMSP平台筛选芎附散的活性成分及靶点;借助STRING平台构建靶蛋白相互作用网络;利用BiNGO和MCODE插件进行靶基因GO生物过程富集和聚类分析;通过DAVID数据库富集分析关键靶点的KEGG信号通路。结果从芎附散中筛选得到丁苯酞、(Z)-藁本内酯、α-香附酮、洋川芎内脂A、洋川芎内脂I等14种活性成分;作用于DRD5、HTR2A、ADR、MAOA、F2、VEGF、NOS3、ESR1、PGR等30个关键靶点;参与内皮细胞与血管形成、多巴胺受体信号通路等12个主要GO生物过程子簇;调控血小板活化、血管内皮生长因子、钙离子、雌激素信号通路,以及5-羟色胺、多巴胺、γ-氨基丁酸等神经递质通路。结论初步解释了芎附散治疗神经系统、心血管系统以及妇科疾病的药效物质基础及其作用机理。

Abstract:Objective Based on the idea and method of network pharmacology,the relationship among active components,targets and pathways of Xiong Fu powder was studied,and its mechanism was discussed.Methods The active components and targets of Xiong Fu powder were screened using BATMAN-TCM database and TCMSP platform,the targets protein-protein interaction network was built using STRING platform,and BiNGO and MCODE plug-ins were used for the enrichment and clustering analysis of target gene GO biological process.KEGG signal pathways of key targets were analyzed through the enrichment of DAVID database.Results Fourteen active components were screened from Xiong Fu powder,including butylphthalide,(Z)-Ligustilide,α-Cyperone,Senkyunolide A,Senkyunolide I,etc.It acts on 30 key targets,include DRD5,HTR2 A,ADR,MAOA,F2,VEGF,NOS3,ESR1 and PGR,etc;and is involved in 12 major GO biological process subclusters,such as regulation of angiogenesis,and dopamine receptor signaling pathway;and regulates platelet activation,vascular endothelial growth factor,calcium ions,estrogen signaling pathways,and neurotransmitter pathways such as Serotonergic synapse,Dopaminergic synapse,GABAergic synapse.Conclusion The effective substances and mechanism of Xiong Fu powder in the treatment of nervous system,cardiovascular system and gynecological diseases were revealed.
