麻黄-桂枝药对抗炎、镇痛作用研究

摘    要:目的观察麻黄-桂枝(Eph-CR)药对的抗炎、镇痛作用。方法将SD大鼠按体质量随机分为对照组,阳性药组,Eph-CR高、中、低剂量组(13.5、6.75、3.375 g·kg^-1),每组8只。采用角叉菜胶致大鼠足肿胀实验及大鼠棉球肉芽肿实验,分别观察各组大鼠足肿胀度及肉芽肿质量,并计算抑制率。将KM小鼠按体质量随机分为对照组,阳性药组,Eph-CR高、中、低剂量组(20、10、5 g·kg^-1),每组10只。采用醋酸致小鼠腹腔毛细血管通透性增加实验、二甲苯致小鼠耳肿胀实验、醋酸扭体实验、热板法及福尔马林实验,分别观察各组小鼠毛细血管通透性、耳肿胀度、扭体次数、痛阈值及Ⅰ、Ⅱ相阶段的舔足时间,并计算抑制率、痛阈提高百分率。结果与对照组比较,Eph-CR高(2~5 h)、中(3、4 h)剂量组大鼠的足肿胀度明显降低(P<0.05);Eph-CR高、中、低剂量组的大鼠肉芽肿质量显著降低(P<0.05,P<0.001)。与对照组比较,Eph-CR高、中、低剂量组小鼠腹腔毛细血管通透性增加显著降低(P<0.05,P<0.01,P<0.001),耳肿胀度显著降低(P<0.01,P<0.001);Eph-CR高、中剂量组小鼠扭体次数显著降低(P<0.05,P<0.01),福尔马林镇痛模型中Ⅱ相阶段小鼠舔足时间明显缩短(P<0.001);Eph-CR高剂量组小鼠热板法实验中60、90、120 min痛阈提高百分率显著增加(P<0.05),福尔马林镇痛模型中Ⅰ相阶段小鼠舔足时间显著减少(P<0.05)。结论 Eph-CR药对对急、慢性炎症均具有明显的抑制作用,高、中剂量具有镇痛的药理活性,而且高剂量还具有一定的中枢镇痛作用。

Abstract:Objective To investigate the anti-inflammatory and analgesic effects of Ephedrae herba-Cinnamomi ramulus(Eph-CR) herb pair.Methods SD rats were randomly divided into control group,positive drug group,and Eph-CR high,medium and low dose groups(13.5,6.75,3.375 g·kg^-1).Carrageenan-induced paw edema and cotton pellet-induced granuloma formation were carried out for observing paw swelling degree and granuloma weight in rats,and the inhibition rate was calculated.KM mice were randomly divided into control group,positive drug group,and Eph-CR high,medium and low dose groups(20,10,5 g·kg^-1).Acetic acid-induced peritoneal permeability increase,xylene-induced ear edema,acetic acid-induced writhing test,hot plate test and formalin test were employed to detect peritoneal permeability,ear swelling degree,number of writhes,pain threshold and licking time in mice;the inhibition rate and the percentage of pain threshold were then calculated.Results Compared with the control group,the paw swelling degree of rats decreased significantly in Eph-CR high dose group at 2,3,4,5 h and medium dose group at 3,4 h(P<0.05).Granuloma weight in rats decreased significantly in Eph-CR high,medium and low dose groups(P<0.05,P<0.001).Compared with the control group,peritoneal permeability decreased significantly(P<0.05,P<0.01,P<0.001),and the ear swelling degree decreased significantly(P<0.01,P<0.001) in mice of Eph-CR high,medium and low dose groups.Number of writhes decreased significantly(P<0.05,P<0.01),and licking time of mice in formalin test at the second phase was reduced significantly(P<0.001) in Eph-CR high and medium dose groups.Pain threshold of mice in hot plate test at 60 min,90 min and 120 min prolonged significantly(P<0.05),and licking time of mice in formalin test at the first phase was reduced significantly(P<0.05) in Eph-CR high dose group.Conclusion Eph-CR herb pair has obvious inhibitory effect on acute and chronic inflammation.Eph-CR at high and medium doses have the pharmacological activity of analgesia,and the high dose group has certain central analgesic effect.
