补肾活血汤改善膝骨关节炎大鼠行为学的实验研究

摘    要:目的观察补肾活血汤对膝骨关节炎大鼠模型行为学的影响。方法将18只SD雄性大鼠随机分为补肾活血汤组、模型组和空白组。补肾活血汤组和模型组大鼠双膝关节腔注射4%木瓜蛋白酶0.2 mL,隔日注射1次,连续注射2周。造模成功后,补肾活血汤组予补肾活血汤中药灌胃给药,模型组和空白组大鼠予以蒸馏水灌胃处理,连续给药4周。比较造模前后各组大鼠的精神状态、体质量、膝关节活动度等。收集各组大鼠双膝关节影像学图像,观察各组大鼠双膝关节面间隙狭窄、关节面磨损及变形、骨赘生成等情况。给药结束后,测定各组大鼠自主活动次数、旷场实验10 min活动总距离和平均速度,以及水迷宫实验找到平台时间和活动距离等。结果造模后,补肾活血汤组、模型组大鼠较空白组大鼠精神萎靡、反应迟钝,双膝关节可见肿胀、活动不利,补肾活血汤组、模型组与空白组体质量比较明显减轻(P<0.05);补肾活血汤组和模型组大鼠双膝关节面间隙狭窄、关节面磨损、变形及骨赘生成较空白组大鼠双膝明显。给药4周后,模型组大鼠较补肾活血汤组、空白组大鼠自主活动次数减少、旷场实验10 min活动总距离和平均速度降低及水迷宫实验找到平台时间和活动距离延长(P<0.05)。结论补肾活血汤具有改善膝骨关节炎SD大鼠行为学指标与治疗膝骨关节炎的功效。

Experimental Study of Bushen Huoxue Decoction on Improving Ethology of Rats with Knee Osteoarthritis

Abstract:Objective:To evaluate the effect of Bushen Huoxue Decoction(BHD)on the ethology of SD rats with knee osteoarthritis.Methods:18 SD male rats were randomly divided into BHD group,the model group and the blank group.BHD group and the model group were given chloral hydrate intraperitoneal anesthesia by injecting papain 0.2 mL with a concentration of 4%into the knee joint cavity.The blank group was anesthetized with chloral hydrate only.Injection was given once every other day for two weeks.After successful modeling,BHD group was given BHD by intragastric administration.The model group and the blank group were treated with distilled water.The drug was administered continuously for 4 weeks.The mental state,body weight and knee joint activity of SD rats were compared before and after modeling.X-ray films were used to collect the imaging images of the knees of SD rats in each group.The narrowing of the knee joint space,the wear and deformation of the knee joint surface and the formation of osteophytes were observed in each group.After administration,the number of spontaneous activities,the total distance and average speed of open field test for 10 minutes,and the time and distance of platform found by water maze experiment were measured.Results:Compared with the blank group,the model group and BHD group were listless and unresponsive with visible swelling of both knees and activity disadvantage,and body weight decreased(P<0.05).The narrowing of the knee joint space,the wear and deformation of the knee joint surface and the formation of osteophytes were more obvious than the blank group.After 4 weeks of administration,compared with BHD group and the blank group,the number of autonomous activities in the model group decreased;the total distance and average speed decreased in open field test for 10 minutes;the time and distance of finding platform in water maze experiment was lengthened(P<0.05).Conclusion:BHD has the effect of improving the behavioral indexes of SD rats with knee osteoarthritis.
