参附强心颗粒中附片减毒增效工艺研究

摘    要:目的优选参附强心颗粒中附片提取工艺。方法以经典原方的水提取工艺为基础,为附片的提取工艺初步设计4种不同的水提取方法,其中,样品1、样品2、样品4分别是水提物、醇沉物以及醇沉后高温处理产物,样品3经水提且高温处理。测定4个样品生物碱含量、小鼠及豚鼠急性毒试验实验研究。结果4个样品6种生物碱总和分别为:0.213、0.163、0.123、0.141 mg/g,以样品3的生物碱含量最低。小鼠毒性试验结果显示,4个附片提取物样品的LD50(95%可信限)分别为152.3(140.9~164.8)、155.0(139.0~172.8)、181.4(172.3~190.9)、150.7(139.4~162.9)g生药/kg,其中样品3的LD50值最大。豚鼠毒性试验结果显示:4种附片提取物样品灌胃豚鼠的半数致心律失常量(AD50)(95%可信限)分别为75.2(65.3~86.4)、73.8(62.8~83.5)、119.8(109.2~130.4)、91.9(82.2~110.4)g生药/kg,LD50(95%可信限)分别为72.6(67.8~77.9)、85.1(78.8~102.6)、109.7(101.6~119.4)、96.0(90.5~101.7)g生药/kg,其中样品3的AD50、LD50值均最大。结论附片经水提且高温处理工艺得到的样品毒性最低,其安全性最高。

Abstract:Objective:To select the extraction process of monkshood in Shenfu Qiangxin Granules.Methods:Based on the water extraction process of the classical original formula,four different water extraction methods were designed for the extraction process of monkshood.Sample 1,sample 2 and sample 4 were water extract,alcohol precipitate and high temperature treatment products after alcohol precipitation respectively.Sample 3 was extracted by water and treated at high temperature.The content of alkaloids in four samples and the acute toxicity test in mice and guinea pigs were determined.Results:Among the four samples,the alkaloid content of sample 3 was the lowest.The results of mouse toxicity test showed that LD50 of sample 3 was the highest.The results of toxicity test on guinea pigs showed that AD50 and LD50 of sample 3 were the highest.Conclusion:After water extraction and high-temperature treatment,the toxicity of monkshood is the lowest and its safety is the highest.
