四君子汤调节2型糖尿病小鼠糖脂代谢紊乱的活性部位筛选研究

摘    要:目的筛选四君子汤调节2型糖尿病(Type 2 diabetes mellitus,T2DM)小鼠糖脂代谢紊乱的活性部位。方法采用高糖高脂饲料喂养联合腹腔注射小剂量(40 mg·kg^-1)链脲佐菌素(STZ)诱导建立T2DM小鼠胰岛素抵抗(Insulin resistance,IR)模型。将雄性ICR小鼠随机分为空白组、模型组、阳性药组(盐酸二甲双胍,0.03 g·kg^-1)、SJZT组(四君子汤水提液,8.48 g·kg^-1)、SJZT-U组(四君子汤水提醇沉液,8.48 g·kg^-1)及SJZT-D组(四君子汤水提醇沉物,8.48 g·kg^-1),每组10只。灌胃给药,每天1次,连续28 d。每周测定小鼠体质量;采用血糖仪检测小鼠空腹血糖(FBG);采用ELISA试剂盒测定血清空腹胰岛素(FINS)水平及肝脏中肝糖原(LG)含量;计算IR指数(HOMA-IR);灌胃葡萄糖溶液(1 g·kg^-1)进行口服葡萄糖耐量(OGTT)检测,计算血糖-时间曲线下面积(AUC);采用生化试剂盒检测血清中总胆固醇(T-CHO)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)及甘油三酯(TG)含量。结果与空白组比较,模型组小鼠体质量、AUC、HOMA-IR及FBG、FINS、T-CHO、TG含量均明显升高(P<0.01),HDL-C、LG含量明显降低(P<0.05,P<0.01)。与模型组比较,各给药组小鼠的AUC、FINS含量明显降低(P<0.05,P<0.01);SJZT及SJZT-D组小鼠的FBG、HOMA-IR明显降低(P<0.01),LG含量明显升高(P<0.01);SJZT及SJZT-U组小鼠的T-CHO含量明显降低(P<0.05);SJZT组小鼠的TG含量明显降低(P<0.05)。结论四君子汤及其不同提取部位能调节T2DM小鼠的糖脂代谢紊乱,其降糖有效部位为水煎液及水提醇沉物,调脂有效部位为水煎液及水提醇沉液。

Abstract:Objective The active parts of Sijunzi decoction regulating glycolipid metabolism disorder in type 2 diabetes mellitus(T2 DM)mice were screened. Methods The insulin resistance model of T2 DM in mice was induced by high glucose and high fat diet combined with intraperitoneal injection of low dose(40 mg·kg^-1)streptozotocin(STZ). ICR mice were randomly divided into blank group,model group,metformin hydrochloride(0.03 g·kg^-1)group,Sijunzi decoction water extract(SJZT,8.48 g·kg^-1)group,Sijunzi decoction water extract alcohol precipitate(SJZT-D, 8.48 g · kg^-1)group and supernatant of Sijunzi decoction water extract alcohol precipitation(SJZT-U,8.48 g·kg^-1)group,10 mice in each group. Drugs were given intragastrically once a day for 4 weeks. Body mass(W)of mice was weighed weekly;fasting blood glucose(FBG)was measured using blood glucose meter,fasting insulin(FINS)level was measured by enzyme linked immunosorbent assay(ELISA),insulin resistance index(HOMA-IR)was calculated. Oral glucose tolerance(OGTT)was measured by oral intake of glucose solution(1 g·kg^-1),and area under blood glucose curve(AUC)was calculated. The contents of total cholesterol(T-CHO),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C)and triglyceride(TG)in serum were measured by biochemical kits,and the content of glycogen in liver(LG)was determined by enzyme-linked immunosorbent assay(ELISA). Results Compared with the blank group,the contents of FBG,FINS,HOMA-IR,T-CHO,and TG,the body mass and AUC in the model group were higher(P < 0.01),the contents of HDL-C and LG decreased significantly(P < 0.05,P < 0.01). Compared with the model group,the AUC and content of FINS in drugs treated groups decreased(P < 0.05,P < 0.01);the content of FBG,and HOMA-IR in SJZT group and SJZT-D group decreased(P < 0.01);and LG contents in creased(P < 0.01). The content of T-CHO decreased in SJZT group and SJZT-U group(P < 0.05);the content of TG in SJZT group was lower than that in model group(P < 0.05).Conclusion Sijunzi decoction can adjust the glucose-lipid metabolism disorder in T2 DM mice. Preliminarily screening showed that the effective parts for the blood sugar-reducing included water decoction and alcohol precipitate of water-extract;and the fat-adjusting effective part is the water decoction and supernatant of waterextract alcohol precipitation solution.
