络风宁1号方对心梗大鼠血管内皮功能的治疗作用

摘    要:目的观察络风宁1号方对心梗大鼠血管内皮功能的治疗作用并探讨其机制。方法选雄性SD大鼠,制作心梗大鼠模型,将术后成活大鼠随机分为假手术组(7只)、模型组(8只)、络风宁1号组(7只)、联合用药组(8只)、卡托普利组(8只),造模后络风宁1号组及联合用药组予含生药量4.81 g/(kg·d)的络风宁1号方汤剂,卡托普利组及联合用药组予卡托普利药液7.5 mg/(kg·d),假手术组及模型组予相同体积蒸馏水,每日2次,连续4周后处死大鼠,检测血管内皮活性物质,TTC染色法测定心肌梗死面积,HE染色观察心肌病理学改变。结果络风宁1号组、联合用药组、卡托普利组大鼠血浆一氧化氮(NO)、前列环素(PGI2)高于模型组(P<0.05),络风宁1号组、联合用药组、卡托普利组大鼠血浆内皮素(ET)、血栓素A2(TXA2)、血浆蛋白C受体(sEPCR)、可溶性血栓调节蛋白(s TM)低于模型组(P<0.05或P<0.01),络风宁1号组与联合用药组NO、PGI2、ET、TXA2含量与卡托普利组比较无明显差异(P>0.05);与卡托普利组比较,联合用药组血清sEPCR、sTM含量降低但无明显差异(P>0.05)。与模型组比较,络风宁1号组、联合用药组、卡托普利组能减小心肌梗死范围(P<0.05或P<0.01);与卡托普利组比较,络风宁1号组、联合用药组梗死范围加大,但差异无统计学意义(P>0.05)。结论络风宁1号方可以改善心梗大鼠血管内皮功能,减小心肌梗死范围。

Abstract:Objective:To study the effect of Luofengning-1 Decoction on vascular endothelial function in rats with myocardial infarction.Methods:The rats were randomly divided into the sham operation group(7 rats),the model group(8 rats),Luofengning-1 Decoction group(7 rats),combination group(8 rats)and captopril group(8 rats).Each group was given corresponding drugs;the sham operation group and the model group were given distilled water of the same volume twice a day.After 4 weeks,the rats were killed.The vascular endothelial active substances were detected;the infarct area was measured by TTC staining,and the myocardial pathological changes were observed by HE staining.Results:NO and PGI2 of Luofengning-1 Decoction group,combination group and captopril group were higher than those of the model group,and there were some significant differences(P<0.01).ET,TXA2,s EPCR and sTM of Luofengning-1 Decoction group,combination group and captopril group were lower than those of the model group,and there were some significant differences(P<0.01 or P<0.05).NO,PGI2,ET and TXA2 in Luofengning-1 Decoction group and combination group were not significantly different from those in captopril group.Compared with the captopril group,the serum sEPCR and sTM levels in the combination group were decreased,but there were no statistical differences.Compared with the model group,Luofengning-1 Decoction group,combination medication group and captopril group reduced the myocardial infarction area,and there was a statistical difference(P<0.01 or P<0.05).Compared with captopril group,the infarct area of Luofengning-1 Decoction group and combination group increased,but there were no statistical differences.Conclusion:Luofengning-1 Decoction can improve the vascular endothelial function.
