基于网络药理学探讨柴胡三参汤治疗缺血性心律失常的作用机制

摘    要:目的:探讨柴胡三参汤治疗缺血性心律失常多靶点、多通路的作用机制。方法:在TCMSP平台筛选柴胡三参汤活性成分,在PubChem和Swiss Target Prediction数据库进行靶标预测;在GeneCards数据库检索缺血性心律失常的靶点;构建韦恩图得到药物与疾病共同靶点;在STRING构建蛋白互作网络图(PPI);在Cytoscape软件得到各靶点的degree值,建立簇状条形图;进行GO生物功能注释和KEGG通路富集分析。结果:本研究共挖掘到柴胡三参汤215种活性成分及1319个作用靶点、疾病靶点120个、药物-疾病共同靶点43个,共同靶点主要富集在221个生物过程、14条信号通路上。结论:柴胡三参汤可能主要是通过调节白蛋白(ALB)、白细胞介素-6(IL-6)、AKT丝氨酸/苏氨酸激酶1(AKT1)、一氧化氮合成酶3(NOS3)、肿瘤坏死因子(TNF)、血管紧张素I转化酶(ACE)、Serpin家族E成员1(SERPINE1)、激肽原1(KNG1)等靶点,调控钙、Toll样受体、细胞凋亡、mTOR、VEGF等信号通路,发挥调控细胞凋亡、抑制炎症反应、免疫调节、恢复心肌血流、清除自由基等作用治疗缺血性心律失常。

Abstract:Objective:To explore the multi-target and multi-pathway action mechanism of Chaihu Sanshen Decoction in treating ischemic arrhythmia.Methods:The active components of Chaihu Sanshen Decoction were screened on the TCMSP platform,and the targets were predicted in the PubChem and Swiss Target Prediction databases.The target of the disease was retrieved in the GeneCards database.The Venn diagram was used to obtain the common targets of drugs and diseases.Protein-protein interaction(PPI)was constructed in STRING,the degree of each target was obtained in Cytoscape,and a cluster bar chart was established.GO biofunctional annotation and KEGG pathway enrichment analysis were performed.Results:A total of 215 active components and 1319 targets in Chaihu Sanshen Decoction and 120 disease-related targets,and 43 drug-disease targets were obtained.The common targets were mainly enriched in 221 biological processes and 14 signal pathways.Conclusion:Chaihu Sanshen Decoction may mainly adjust albumin(ALB),interleukin-6(IL-6),AKT serine/threonine kinase 1(AKT1),nitric oxide synthase 3(NOS3),tumor necrosis factor(TNF),angiotensin I converting enzyme(ACE),serpin family E member 1(SERPINE1)and kininogen 1(KNG1),and regulate calcium signaling pathway,Toll-like receptor signaling pathway,apoptosis,mTOR signaling pathway,VEGF signaling pathway,so as to regulate apoptosis,inhibit inflammatory response,adjust immune,restore myocardial blood flow,scavenge free radicals to treat ischemic arrhythmias.
