针康法对缺氧缺血性脑损伤新生大鼠细胞凋亡相关因子表达的影响

摘    要:目的观察针康法对缺氧缺血性新生大鼠大脑皮质内细胞凋亡诱导因子及死亡基因受体mRNA表达及细胞凋亡的影响。方法随机将实验用Wistar大鼠分为假手术组、模型组、针刺组、康复组及针康组,又将各组于12、24、48、72 h分为4个亚组(n=8)。后应用Rice-Vannucci法建立缺氧缺血性脑损伤模型。其中假手术组不予任何治疗;模型组在造模成功后只予以同等条件抓取;针刺组进行头穴丛刺,留针2 h;康复组进行转笼转棒训练,每日10 min;针康组针刺方案同针刺组,康复方案同康复组。结果与假手术相比,其余各组的AIF、FasmRNA在各时间点的表达及细胞凋亡情况,差异具有统计学意义(P<0.05);与模型组相比,针刺组、康复组及针康组在各时间点的AIF、FasmRNA表达及细胞凋亡情况差异也有统计学意义(P<0.05);针康组与针刺组及康复组比较,在各时间点的表达,差异有统计学意义(P<0.05)。模型组、针刺组、康复组、针康组在各时间点内的细胞凋亡数与假手术组比较差异具有统计学意义(P<0.05);针刺组、康复组、针康组在各时间点与模型组相比,其细胞凋亡数明显减少,...

Abstract:Objective To observe the effects of acupuncture-rehabilitation therapy on the expression of apoptosisinducing factor,death gene receptor mRNA and apoptosis in cerebral cortex of neonatal rats with hypoxia-ischemia.Methods Wistar rats were randomly divided into sham operation group,model group,acupuncture group,rehabilitation group and needle Kang group.The groups were divided into 4 subgroups at 12 h,24 h,48 h and 72 h(n=8).The model of hypoxic-ischemic brain injury was established by the Rice-Vannucci method.The sham operation group did not receive any treatment;the model group was only grasped under the same conditions after successful modeling;the acupuncture group performed the head acupuncture and the needle was kept for 2 hours;the rehabilitation group was trained to rotate the rod for 10 minutes each;The acupuncture-rehabilitation group acupuncture program is the same as the acupuncture group,and the rehabilitation program is the same as the rehabilitation group.Results Compared with sham operation,the expression of AIF and Fas mRNA in different groups at different time points and apoptosis were significantly different(P<0.05).Compared with the model group,acupuncture group,rehabilitation group and needle There were significant differences in AIF,Fas mRNA expression and apoptosis between the Kang group at each time point(P<0.05).There was a difference in the expression of the Acupuncture group and the rehabilitation group at each time point.Statistical significance(P<0.05).Compared with the sham operation group,the number of apoptosis in the other groups was significantly different at each time point(P<0.05);the number of apoptosis in the latter three groups was significantly reduced compared with the model group at each time point.There was significant(P<0.05);and the number of apoptosis in the needle group was statistically significant compared with the acupuncture group and the rehabilitation group(P<0.05).Conclusion Acupuncture-rehabilitation therapy can exert brain protective effects by inhibiting the expression of apoptosis-inducing factor and death gene receptor mRNA in the cerebral cortex and inhibiting apoptosis after hypoxic-ischemic brain damage.
