两种慢性萎缩性胃炎大鼠模型的建立与比较

摘    要:目的比较幽门弹簧植入术配合高盐热淀粉糊灌胃法及MNNG复合多因素法建立慢性萎缩性胃炎大鼠模型的生物学特性及稳定性。方法分别采用幽门弹簧植入术配合高盐热淀粉糊灌胃法和MNNG复合多因素法建立慢性萎缩性胃炎大鼠模型,记录其体质量、进食量、饮水量,应用HE染色评价胃黏膜病理损伤,Elisa评价血清PGⅠ/PGⅡ和GAS表达水平,对两种模型进行综合评价。结果两种模型在造模过程中大鼠均出现体质量降低,进食及饮水量较空白组减少;病理方面MNNG模型组大鼠胃黏膜可见明显肠上皮化生及异型增生,幽门弹簧植入术淋巴滤泡较多,炎症反应明显;血清学方面MNNG模型组PGⅠ/PGⅡ含量较空白组下降(P <0.05),MNNG模型组及手术模型组GAS含量均较正常组显著降低(P <0.01)。结论幽门弹簧植入术诱导的CAG模型大鼠主要萎缩部位在胃窦,以炎症表现为主;MNNG诱导的CAG大鼠模型萎缩部位在全胃,更趋向于萎缩性胃炎癌前病变的病理表现。

Abstract:Objective To compare the biological characteristics and stability of pyloric spring implantation combined with high-salt hot starch paste gavage and MNNG compound multifactor method in the establishment of chronic atrophic gastritis rat model. Methods Established two different CAG models in rats, one was induced by implantation of a pyloric spring and intragastic administration of hot starch paste with high-salt, the other was induced by MNNG. Compared the two models from pathological conditions, the level of serum gastrin, serum PGⅠ/PGⅡ and evaluated the characteristics of the two models. Results Weight loss occurred in both models in rats during the process of modeling, the intake of food and water was less than that of the blank group. The model established by MNNG could lead to intestinal metaplasia. The model established by Pylorus spring implantation with high salt hot starch paste showed inflammatory cell infiltration, lymphoid follicular formation. Compared with the control group, PGⅠ/PGⅡ level was reduced in MNNG group(P<0.05), two models of serum gastrin hormone(GAS) were significantly reduced(P<0.01). Conclusion The CAG model reduced by pylorus spring implantation with high salt hot starch paste is given priority to inflammation, and the model reduced by MNNG compound multi factors appeared metaplasia of intestinal epithelium.
