摘    要:目的建立HPLC法同时测定骨折挫伤胶囊中羟基红花黄色素A、血竭素、芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚的含量。方法采用ZORBAX Eclipse Plus-C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%磷酸水溶液,梯度洗脱,流速:1.0 mL·min^-1,柱温:35℃,检测波长:403、440、254 nm。结果羟基红花黄色素A、血竭素、芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚质量浓度分别在2.111~21.11μg·mL^-1(r=0.999 1)、1.483~14.83μg·mL^-1(r=0.999 3)、1.180~11.80μg·mL^-1(r=0.999 7)、0.8182~8.182μg·mL^-1(r=0.999 7)、1.011~10.11μg·mL^-1(r=0.999 7)、1.075~10.75μg·mL^-1(r=0.999 6)和1.045~10.45μg·mL^-1(r=0.999 7)范围内与峰面积呈良好的线性关系;平均回收率(n=6)分别为100.30%(RSD=2.24%)、97.42%(RSD=1.63%)、96.21%(RSD=0.92%)、101.17%(RSD=2.74%)、95.84%(RSD=1.31%)、97.71%(RSD=2.49%)和95.50%(RSD=1.15%)。结论该方法准确度高、重复性好,可为骨折挫伤胶囊的质量控制提供参考。

Abstract:Objective To establish an HPLC method for simultaneous determination of hydroxysafflor yellow A,dracorhodin,aloe-emodin,rhein,emodin,chrysophanol and rhyscion in Guzhe Cuoshang capsules.Methods The analysis of 3% acetonitrile methanol extract of this drug was performed on ZORBAX Eclipse Plus-C18 column(250 mm×4.6 mm,5μm) by gradient elution of acetonitrile-0.2% phosphoric acid solution at a flow rate of 1.0 mL·min^-1.The column temperature was 35℃and the detection wavelength was set at 403 nm,440 nm,254 nm.Results Good linear relationships were found for hydroxysafflor yellow A,dracorhodin,aloe-emodin,rhein,emodin,chrysophanol and physcion in the ranges of 2.111-21.11 μg·mL^-1(r=0.999 1),1.483-14.83 μg·mL^-1(r=0.999 3),1.180-11.80μg·mL^-1(r=0.999 7),0.818 2-8.182 μg·mL^-1(r=0.999 7),1.011-10.11 μg·mL^-1(r=0.999 7),1.075-10.75μg·mL^-1(r=0.999 6),and 1.045-10.45μg·mL^-1(r=0.999 7),respectively.The average recoveries(n=6) were 100.30%(RSD=2.24%),97.42%(RSD=1.63%),96.21%(RSD=0.92%),101.17%(RSD=2.74%),95.84%(RSD=1.31%),97.71%(RSD=2.49%) and 95.50%(RSD=1.15%),respectively.Conclusion The method is accurate and reproducible,and can provide reference for the quality control of Guzhe Cuoshang capsules.
