泻心汤有效成分对高糖诱导的HK-2细胞迁移的抑制作用

摘    要:目的研究泻心汤有效成分对高糖诱导的肾小管上皮(HK-2)细胞迁移作用的影响。方法体外高糖(30mmol·L-1)培养HK-2细胞,分别加入不同浓度的大黄酸、小檗碱和黄芩苷,孵育48h后采用Transwell法检测细胞迁移能力,荧光定量PCR法检测黏着斑激酶(FAK)mRNA表达,Westernblot法检测a-平滑肌肌动蛋白(a-SMA)表达;孵育24h后收集细胞上清,利用比色法检测丙二醛(MDA)的含量及超氧化物歧化酶(SOD)活力。结果高糖诱导IIK-2细胞迁移能力增强,大黄酸、小檗碱和黄芩苷均可不同程度地抑制细胞迁移能力,降低FAKmRNA表达、a-SMA蛋白表达和MDA产生,并提高SOD的酶活力。结论泻心汤有效成分具有抑制高糖诱导的HK-2细胞迁移作用。

Abstract:AIM To investigate the effects of active ingredients of Xiexin decoction (Rhein, Berberine and Baicalin) on the cell-migration in human kidney proximal tubular epithelial (HK-2) cells induced by high glucose. METHODS HK-2 cells were cultured by high glucose and treated with different concentrations of Rhein, Berberine and Baicalin. After 48 h, cells migration were measured by the Transwell experiment, and the expressions of focal adhesion kinase(FAK) mRNA were detected by real-time PCR, the expressions of u-SMA were detected by Western blot. After 24 h ,malondiadehyde(MDA) activities were measured by thiobarbituric acid method and SOD activities were measured by xanthine oxidase method. RESULTS Cell-migration was enhanced by high glucose in HK-2 cells. Rhein, Berherine and Baicalin could inhibit cell-migration, the expression of FAK mRNA and the expression of a-SMA, and reduce MDA lev- el, elevate SOD activities. CONCLUSION The active ingredients of Xiexin decoction could inhibite high glucose-in- duced migration of HK-2 cells.
