摘    要:目的:研究熊果酸对巨噬细胞-肝癌细胞共培养体中肝癌细胞迁移和侵袭能力的影响,并初步探讨其作用 机制。方法:采用Transwell迁移和侵袭试验检测熊果酸处理与非处理的巨噬细胞-肝癌细胞共培养体中肝癌细胞的迁 移和侵袭能力的变化;Western印迹法检测共培养体中肝癌细胞上皮-间质转化蛋白E-cadherin,N-cadherin和vimentin的 表达情况。结果:共培养后的肝癌细胞的迁移和侵袭能力显著增强,并发生了上皮-间质样表型转变,上皮-间质转化 的标志蛋白E-cadherin的表达下调,而N-cadherin和vimentin的表达均上调;熊果酸处理的巨噬细胞-肝癌细胞共培养体 中,肝癌细胞的迁移和侵袭能力降低,E-cadherin表达上调,N-cadherin和vimentin的表达下调。结论:熊果酸能够显 著抑制共培养体中肝癌细胞的迁移和侵袭能力及上皮-间质转化能力。

Abstract:Objective: To investigate the eff ect of ursolic acid on the invasion and migration of hepatocellular carcinoma (HCC) cells co-cultured with macrophages, and to explore the underlying mechanisms. Methods: Th e migration and invasion ability of HCC cells in the co-culture system with or without ursolic acid intervention were evaluated by transwell assay. Th e levels of epithelial-mesenchymal transition (EMT) markers E-cadherin, N-cadherin, and vimentin in HCC cells co-cultured with macrophages were detected by Western blot. Results: The migration and invasion ability and EMT were significantly enhanced when co-cultured with macrophages, and the expression of E-cadherin was significantly increased while N-cadherin and vimentin levels were significantly decreased. However, after ursolic acid treatment, the migration and invasion ability were significantly reduced, and the expression of E-cadherin was increased while N-cadherin and vimentin levels were decreased. Conclusion: Ursolic acid exerts inhibitory effect on the ability of migration, invasion, and EMT for HCC, which are enhanced by co-culturing with macrophages.
