CD117、C-erbB-2蛋白在原发性肝细胞性肝癌中的表达及意义

目的　探讨原发性肝细胞肝癌(HCC)中 CD117 及 C- erbB- 2 蛋白的表达特点及其与临床病理的关系,为今后可能的分子靶向治疗提供实验依据。方法　应用免疫组化 SP法,检测 103 例HCC中CD117及C- erbB -2蛋白的表达,对照组为正常肝组织 10 例,肝硬化组织 40 例。结果　CD117在HCC组、肝硬化组和正常肝组织表达阳性率分别为36.89%、37.5%和30%,CD117表达在三组间无显著性差异,但与HCC分期及组织学分级显著相关(P<0.05)。C -erbB- 2 在 HCC组、肝硬化组和正常肝组织表达阳性率分别为 50. 49%、70%和 40%,三组之间存在显著性差异(P <0.05),C erbB 2表达与组织学分级无关,但与临床分期密切相关。CD117 与 C -erbB- 2 共同表达率为12.6%,且两者表达明显相关。结论　CD117 与 C -erbB- 2 在 HCC发生、发展过程中具有重要作用,对于选择性病例可以应用针对CD117和C -erbB- 2的分子靶向药物治疗,还有待进一步研究。

Expression of CD117 and C-erbB-2 in hepatocellular carcinoma and their significance

Objective To explore the expression of CD117 and C-erbB-2 in hepatocellular carcinoma (HCC) and its clinical significance. Methods The expression of CD117 and C-erbB-2 were evaluated by SP immunohistochemical staining in 103 cases of HCC and 40 cases of liver cirrhotic specimens, as well as 10 cases of normal liver specimens. Results The positive expression of CD117 was found in 36.89% of HCC, 37.5% of cirrhostic liver specimens, and 30% of normal liver specimens, respectively. There was no significant difference among the three groups, but the CD117 expression was significant associated with the histological grades and clinical stages. The positive expression of C-erbB-2 was 50.49%, 70% and 40% in HCC, cirrhotic liver specimens and normal liver tissues, respectively. Moreover, statistical difference was found in the expression of C-erbB-2 among three groups. The C-erbB-2 expression was not related to histological grades, but dramatically related to the clinical stages. CD117 expression was associated with the C-erbB-2 expression, and their co-expression was observed in 12.6% of HCCs. Conclusions CD117 and C-erbB-2 may play important role in liver carcinogenesis and the progression of HCC.
