当归多糖对小鼠骨髓基质细胞增殖、细胞周期及细胞凋亡的影响

摘    要:目的研究当归多糖(APS)对小鼠骨髓基质细胞(BMSC)增殖及细胞周期和细胞凋亡的影响。方法原代培养BMSC,随机分为对照组和药物组,作用不同时间后观察APS浓度为50mg/L、100mg/L时对小鼠BMSC80%贴壁时间的影响,MTT法检测APS浓度分别为25mg/L、50mg/L、100mg/L、200mg/L、300mg/L时对小鼠BMSC增殖的影响;流式细胞仪检测APS浓度分别为25mg/L、50mg/L、100mg/L、200mg/L、300mg/L时对小鼠BMSC细胞周期和细胞凋亡的影响。结果APS作用后的BMSC达80%贴壁所需时间比对照组明显缩短(P〈0.05);在同一作用时间下,各APS浓度组BMSC的增殖能力较对照组增强(P〈0.05),其中50mg/L组和100mg/L组中BMSC增殖能力较其他各组明显增强(P〈0.05);各药物组小鼠BMSC3.5d后各组S期和S+G2/M期均较对照组明显增多(P〈0.05);而G0/G1期细胞均较对照组减少(P〈0.05);50mg/L组和100mg/L组BMSCS期细胞较其它药物组S期细胞数明显增多(P〈0.05);除50mg/L组外,其余各药物组G2/M期细胞较对照组增多(P〈0.05)。各药物组BMSC的凋亡较对照组降低(P〈0.05),并且浓度为100mg/L时其凋亡较其它各组明显减少(P〈0.05)。结论APS可改变BMSC细胞周期,减少凋亡细胞数目进而促进BMSC的增殖。
