乳腺癌妇女外周血 MDSCs 的表达意义

摘    要:目的:探究髓样抑制细胞( Myeloid-derived suppressor cells ,MDSCs)在乳腺癌妇女外周血中的表达意义。方法运用流式细胞术,采用多种荧光抗体( CD11 b、CD14、CD33等)鉴定术,按其疾病类型将所有患者分为实验A组(43例乳腺癌妇女)、实验B组(37例乳腺良性肿瘤妇女),另选取38例身体健康状况良好的妇女作为对照组。外周血中MDSCs的多种表面标志,检测并比较3组妇女外周血中MDSCs以及不同细胞表面标志( CD11 b+CD14-细胞、CD33+CD14-细胞、CD11 b+CD33+CD14-细胞等)水平;记录乳腺癌患者的肿瘤分期情况、淋巴结转移情况、孕激素受体以及雌激素受体表达情况。结果3组外周血中MDSCs水平有差异,实验A组>实验B组>对照组,差异均有统计学意义(P<0.05);实验A组的CD11b+CD14-细胞、CD33+CD14-细胞、CD11b+CD33+CD14-细胞表面标志水平均显著高于实验B组与对照组( P<0.05)。不同临床病理特征的乳腺癌患者间 CD11 b+CD33+CD14-细胞水平差异无统计学意义( P>0.05)。结论乳腺癌妇女外周血的CD11 b+CD33+CD14-细胞表面标志水平、MDSCs水平显著增高,可以作为乳腺癌诊断的生物标志物。

Increased level of MDSCs in the peripheral blood as a biomarker for assessing the development of breast cancer in women
Abstract:Objective To explore the role of peripheral blood MDSCs ( Myeloid-derived suppressor cells ) in assessing the development of breast cancer in women .Method 43 women bearing breast cancer and 37 women bearing benign breast tumor were firstly chosen as two experimental groups , named group A and group B respectively , while 38 healthy women were selected as the control group .Flow cytometry and fluorescent antibodies ( CD11b, CD14, CD33, etc.) were employed to identify multiple surface markers of the peripheral blood MDSCs of the three groups women .The level of peripheral blood of MDSCs and the different cell surface signs (CD11b+CD14-cells, CD33+CD14-cells, CD11b+CD33+CD14-cells etc.) of three groups were then measured and compared .The tumor stage , lymph node metastasis , progesterone receptor and estrogen receptor expression in patients bearing breast cancer were checked .Findings The peripheral blood MDSCs levels of group A , group B and the control group were different , with that of group A higher than that of group B and that of group B higher than that of the control group.The differences were statistically significant (P<0.05).The levels of CD11b+CD14-, CD33+CD14-and CD11b+CD33+CD14-cells of group A were significantly higher than those of group B and those of the control group.However, no significant differences were identified among the three groups in the levels of CD 11b+CD33+CD14-cells.Conclusion Significantly increased levels of CD 11b+CD33+CD14-cells and MDSCs in the peripheral blood may serve as a useful diagnostic marker for assessing the development of breast cancer in women .
