车前子对腹泻模型大鼠小肠组织中水通道蛋白的影响

摘    要:目的观察车前子对腹泻模型大鼠小肠组织中水通道蛋白1(AQP1)、水通道蛋白3(AQP3)的基因和蛋白表达的影响,探讨其治疗腹泻的作用机制。方法将60只SD雄性大鼠采用随机数字表法分为空白组,模型组,阳性对照(氢氯噻嗪)组,车前子低、中、高剂量组,每组10只。除空白组外,其余各组于每日上午予番泻叶药液灌胃复制大鼠腹泻模型,空白组予等量蒸馏水灌胃;每日下午各药物组予相应药物灌胃,空白组和模型组予等量蒸馏水灌胃,连续灌胃14 d。给药期间观察各组大鼠的排便情况。灌胃结束后,在麻醉状态下剖取各组大鼠的小肠组织,采用实时荧光定量PCR法和蛋白免疫印迹法检测小肠组织中AQP1、AQP3的基因和蛋白表达水平。结果模型组大鼠全部排水样稀便;与空白组比较,模型组大鼠小肠组织中AQP1 mRNA、AQP3 mRNA和相应蛋白的表达水平均明显降低,差异有统计学意义(P <0.01)。与模型组比较,车前子各剂量组大鼠粪便基本成形;小肠组织中AQP1 mRNA、AQP3 mRNA和相应蛋白的表达水平均明显升高,差异有统计学意义(P<0.01)。结论车前子可通过上调小肠组织中AQP1、AQP3的基因和...

Abstract:Objective To explore mechanism of action of Plantago asiatica L.in treating diarrhea by observing the effects on the genes and proteins expressions of aquaporin(AQP)1 and AQP3 in small intestinal tissues of diarrhea model rats.Methods Sixty SD male rats were divided into blank group,model group,positive control(hydrochlorothiazide)group,Plantago asiatica L.low,medium and high dose group,with 10 rats in each.Except the blank group,the other groups were given Folium Sennae liquid medicine through administration by gavage to replicate diarrhea model rats every morning,the blank group was given equal amount of distilled water through administration by gavage;each medicine group was given corresponding medicines by gavage,the blank group and model group was given equal amount of distilled water by gavage every afternoon,consecutive gavage for 14 days.The defecation of rats in each group was observed during the administration.After gavage,the small intestinal tissues of rats in each group were dissected under anesthesia,and the genes and protein expression levels of AQP1 and AQP3 in the small intestinal tissues were detected by real-time quantitative PCR and Western blotting.Results The rats in the model group made the passage of loose and liquid stools;compared with the blank group,the expression levels of AQP1 mRNA,AQP3 mRNA and corresponding proteins in the small intestinal tissues of rats in the model group were significantly reduced,and the difference was statistically significant(P<0.01).Compared with the model group,the stools were basically formed in each group of Plantago asiatica L.;the expression levels of AQP1 mRNA,AQP3 mRNA and corresponding proteins in the small intestinal tissues significantly increased,and the difference was statistically significant(P<0.01).Conclusion By upregulating the gene and protein expression levels of AQP1 and AQP3,Plantago asiatica L.can increase the water absorption in the small intestine and regulate water metabolism,achieve the purpose of stopping diarrhea thereby.
