Abstract:Objective To investigate the effect of Shh protein on microRNA-210 expression in cerebral cortex of mice with cerebral infarction. Method A cerebral cortex infarction mouse model was established. The differentially expressed genes in the cerebral cortex of the mice with cerebral infarction were screened by gene chip technology, and the differentially expressed genes were verified by qRT-PCR. After treatment with Shh protein, qRTPCR was used to observe the expression of Shh protein on microRNA-210 in cerebral cortex of mice with cerebral infarction. ATP kit was used to detect the changes of Shh protein on ATP activity in cerebral cortex of mice with cerebral infarction. Results Compared with the Sham group, 17 microRNA differentially expressed in the cerebral cortex of the cerebral infarction mice were screened by gene chip, among which 8 microRNA were up-regulated and 9 microRNA were down-regulated. MicroRNA-210 was further screened and qRT-PCR was used to verify its upregulated expression in cerebral cortex after cerebral infarction(P< 0.05). Compared with the cerebral infarction group, microRNA-210 expression in the cortical tissue of mice was up-regulated after Shh protein intervention(P< 0.05). Compared with the Sham group, the ATP content in the cerebral infarction group decreased, while increased after Shh protein treatment(P< 0.05). Conclusion Shh protein may play a therapeutic role in cerebral infarction by upregulating microRNA-210 to reverse energy generation.
