结直肠癌缺陷基因蛋白在大肠肿瘤中的表达及临床意义

目的：探讨结直肠癌缺陷基因在大肠腺瘤－癌演序中的蛋白表达及其临床意义。方法：以结直肠癌缺陷基因蛋白胞内部分独特氨基酸链的单抗15041A为一抗，免疫组化ABC法检测21例大肠粘膜、40例伴不同程度不典型增生大肠腺瘤、21例大肠癌组织的蛋白表达情况。结果：结直怕癌缺陷基因蛋白表达率，大肠粘膜及轻度不典型增生大肠腺瘤100％，中度不典型增生腺瘤93％，明显高于重度不典型增生腺瘤（45％）及大肠瘤（33％），P＜0．05。结论：结直肠癌缺陷基因蛋白表达缺失与大肠腺瘤演变及大肠癌发展相关，免疫组化法检测结直肠癌缺陷基因蛋白发现癌前病变，研究大肠瘤演变及大肠癌发展具有重要意义。

Protein Expression of DCC Genein Colorectal Tumor and Its Clinical Significance

Objective:To study the protein expression of DCC gene in colorectal polyps carcinoma. and its significance in the development of colorectal adenoma carcinoma sequence.Methods:The samples of normal colorectal mucosa of 21 cases persons, atypical proliferative colorectal adenoma of 40 cases, and colorectal carcinoma of 21 cases were colected for immunohistochemical analysis. The expression of DCC protein was investigated with monoclonal antibody (15041 A).Results: In normal colorectal mucosa and mild atypical proliferative adenomas the positive expression rate of DCC protein was 100%. Which were significantly higher than that in moderate atypical proliferative adenoma (45%) and carcinoma tissues (33%). The DCC protein expression was declined with the development from colorectal adenoma carcinom. Conclusion:Diminished expression DCC protein is a useful marker in predicting of colorectal tumor, and the reductive DCC expression is related to oncogenesis and development of colorectal cancer.
