日本血吸虫感染家兔红细胞上非游离性循环抗原的分离及动态水平研究

摘    要:采用特定方法成功地从50和100条尾蚴感染的家兔红细胞上分离出非游离性循环抗原(NF-CAg),并应用抗SEA和AWA的多克隆及单克隆抗体进行ELISA,对NF-CAg水平的动态进行了观察。结果表明,NF-CAg的水平在感染后8~10wk间达高峰,10wk后下降到较低水平。提示检测NF-CAg的优势是在感染早期(10wk内)。结果还显示,NF-CAg水平与感染程度密切相关。在检测NF-CAg时,特异性的多克隆抗体具有与单克隆抗体相似的检测效果。本文为探索血吸虫循环抗原的新途径和新内容及其归宿问题,提供了重要依据。

Abstract:Non-free circulating antigen(NF-CAg) on red blood cells of rabbits infected with 50 or 100 cercariae of Schistosoma japonicum was successfully separated by a defined method. The separated NF-CAg was detected using ELISA (indirect method) with polyclonal antibody (PcAb) or monoclonal antibody (McAb)against soluble egg antigen(SEA)and polyclonal antibody against adult worm antigen(AWA )at various periods after infection, The results showed that the peak level of NFD-CAg appeared at the 8th and 10th week after infection. The superiority of NF-CAg detection was its applicability at the early stage of the infection (within ten weeks). The level of NF-CAg was closely related to the intensity and duration of the infection. The major components of NF-CAg, the combined CAgs of worm origin and egg origin, were determined, The detection effects of specific NF-CAg by ELISA with anti-SWA PcAb and anti-AWA PcAb were similar to those by ELISA with anti-SEA McAb. This study provided important bases for approaching new ways of studying N F-CAg and understanding the outcome of free schistosome circulating antigen ( F-CAg)in the blood circulation of infected hosts.
