EAE豚鼠T细胞平衡在NPY干预下的研究

目的探讨神经肽Y（NPY）对EAE豚鼠发病及对Th1/Th2细胞平衡的影响,探索NPY对EAE发病影响的免疫调节机制。方法 30只豚鼠随机分为正常对照组、EAE对照组、NPY干预组。观察三组豚鼠血清IL-4、IFN-γ的含量,Th1/Th2比值,以及发病情况。结果 EAE豚鼠发病后血清IL-4含量降低,血清IFN-γ含量升高,Th1/Th2比值升高,Th1/Th2细胞失衡;而NPY干预组高峰期血清IL-4含量明显增高（P〈0.001）,血清IFN-γ含量明显降低（P〈0.001）,Th1/Th2比值降低（P〈0.001）,一定程度地纠正Th1/Th2细胞失衡。结论侧脑室注射NPY对EAE能起到保护作用,其保护机制可能是通过调节Th1/Th2细胞平衡,促进IL-4分泌,抑制IFN-γ的产生而发挥。

Regulation of NPY on the Th1/Th2 balancement of Experimental Allergic Encephalomyelitis in Guinea pig

Objective To investigate the effect of neuropeptide Y on the Th1/Th2 balancement of EAE,to investigate the immune regulation of NPY on EAE.Method 30 Guinea pigs were assigned randomly to three groups： the normol,the EAE control,and the NPY interference group.The levels of cytokines IL-4 and IFN-γ in blood serum were detected.Results The level of IL-4 in blood serum were lower,the ratio of Th1/Th2 were increased.Comparing to the EAE control group,the levels of IFN-γin the blood serum of the NPY interference group were significantly higher,while the level of IL-4 in blood serum of the NPY interference group was significantly lower.The ratio of Th1/Th2 were decreased,the disturbed balancement were corrected.Conclusion The protected effect of NPY on EAE maybe carried out through the regulation on the Th1/Th2 balancement,the increased level of IL-4 and the decreased level of IFN-γ.
