锂盐对小鼠吗啡行为敏化的影响

摘    要:目的探讨锂盐对小鼠吗啡行为敏化的影响。方法测定小鼠的自主活动 ,观察锂盐对小鼠自主活动的影响。急性吗啡 ( 10mg/kg)处理小鼠前给予锂盐 ,观察锂盐对急性吗啡引起小鼠自主活动的影响。慢性吗啡处理小鼠 ,建立吗啡行为敏化小鼠模型 ,观察锂盐对吗啡行为敏化的影响。结果 1.锂盐 ( 2~8mEq/kg)明显抑制小鼠的自主活动 ,并呈剂量依赖性。 2 .锂盐剂量依赖性地抑制急性吗啡所致小鼠的高活动性。 3 .锂盐剂量依赖性地阻断小鼠吗啡行为敏化的表达 ,但不影响行为敏化的形成。结论锂盐对中枢神经系统具有抑制作用 ,并能够抑制吗啡引起的小鼠高活动性以及行为敏化的表达 ,提示锂盐可能对吗啡的精神依赖性具有干预作用。

Abstract:LI Jun-xu,LIANG Jian-hui National Institute on Drug Dependence, Peking University, Beijing 100083,China Objective To investigate the effects of lithium salt on morphine-induced behavioral sensitization. Methods Locomotor activity was measured every 5 minutes for 1 hour immediately after administration with lithium in mice. Behavioral sensitization to morphine was induced by intraperitoneal injection of 10mg/kg morphine once daily for 7days in mice, and treatment with the combination of morphine (i.p.) and lithium (i.p.) to investigate the effects of lithium on the sensitization to morphine. Results 1. Lithium dose-dependently inhibited the locomotor activity in mice. 2. Lithium significantly blocked the expression of morphine behavioral sensitization in mice, but did not affect the development of behavioral sensitization. ConclusionsLithium has inhibitory effects on the CNS by multiple mechanisms and the non-specific inhibitory effect may suppress the behavior of morphine sensitization in mice. These findings indicate that lithium might have the clinical potential to treat morphine addiction. [
