STCl过表达对宫颈癌CaSki细胞生物学行为的影响

摘    要:【目的】研究转染STCl基因对人宫颈癌CaSki细胞生物学行为的影响。【方法】通过MTT、集落形成、划痕实验、Tranwell实验、裸鼠成瘤等方法检测STCl基因转染对宫颈癌CaSki细胞增殖、迁移、侵袭、体内成瘤能力的影响。【结果1MTT和集落形成实验表明,与空载体组相比,CaSki/STCl细胞生长受到抑制,细胞存活率和集落形成率较低(P〈0.05)。划痕实验显示CaSki/STCl较CaSki/NC组细胞迁移率降低(35±3.2% VS 64±4.5%,P〈0.05)。TranweⅡ实验显示CaSki/NC的85±5,而CaSki/STCl组穿膜细胞数较少为54±4(P〈0.05)。裸鼠成瘤实验显示,CaSki/STCl组和CaSki/NC组的肿瘤平均质量依次为(O.285士0.057)g、(0.523±0.154)g,差异具有统计学意义(P,0.01)。【结论】sTCl过表达抑制宫颈癌细胞增殖、转移及侵袭,STCl基因可用于宫颈癌的基因治疗。

Effect of Overexpression of STC1 on Biological Behavior in Human Cervical Cancer Cell Line CaSki

Abstract:[Objective] To investigate the effect of STC1 gene transfection on the biological behavior in hu- man cervical cancer cell line CaSki. [Methods] MTT, colony conforming assay, scrape experiment, Transwell assay and tumorigenicity testing were used to evaluated the effect of STC1 overexpression on the proliferation, migration, invasion and tumor formation of human cervical cancer cell line CaSki. [Results]MTT and colony conforming assay indicated that the cell growth was inhibited and the cell survival and colony conforming rate were lower in CaSki/STC1 group compared with CaSki/NC group( P 〈0.05). The scrape experiment showed that the cell migration decreased in CaSki/STC1 group compared with CaSki/NC group(35 ±3.2% vs 64 ± 4.5%, P 〈0.05). Tranwell assay showed that the number of transmembrane cells in CaSki/NC group was 85±5 which was higher than that in CaSki/STC1 group(54±4)( P 〈0.05). Tumorigenicity testing showed that the average weight of tumor in CaSki/STC1 group and CaSki/NC group were 0. 285±0. 057g and 0. 523± 0. 154g, respectively, and there was significant difference between two groups( P 〈0.01). [ConclusionlSTC1 overexpression can inhibit the proliferation, invasion and metastasis of cervical cancer cells. STC1 gene can be used for the gene therapy of cervical cancer.
