不同抗氧化活性蔬菜汁对游泳诱导的氧化应激的干预作用

摘    要:目的:观察藕汁、油菜汁、黄瓜汁对游泳大鼠抗氧化功能的作用,探讨其可能机制。方法:成年雄性Wistar大鼠50只,随机分为安静对照组、游泳运动组、藕汁+游泳组、油菜汁+游泳组和黄瓜汁+游泳组,每组10只。前两组每天灌胃蒸馏水5 ml,后三组每天分别灌胃新鲜藕、油菜、黄瓜榨汁5 ml,共4周。实验结束时,游泳运动组和三个蔬菜汁组进行一次6 h吊尾游泳,游泳后即刻采血,取心、肝和骨骼肌,安静对照组除不游泳外其它处理均相同。测定心脏、肝脏、骨骼肌组织及血清MDA含量和总抗氧化力;血清维生素C和E,SOD、GSH-Px、CAT活性,GSH含量;骨骼肌线粒体膜电位、膜流动性。结果:与安静对照组比较,游泳运动组血清、心脏、肝脏与骨骼肌组织及骨骼肌线粒体MDA含量显著升高;血清VC、VE和GSH水平,SOD、GSH-Px和CAT活性,血清和组织总抗氧化力显著降低;腓肠肌线粒体的膜电位显著降低,膜流动性显著下降。与游泳运动组比较,三个蔬菜汁组血清、心脏、肝脏与骨骼肌线粒体MDA含量显著减少,总抗氧化力增强,血清VC和GSH含量、SOD、GSH-Px、CAT活性显著增加,心脏和肝脏总抗氧化力显著增强,藕汁组骨骼肌总抗氧化力显著增强。与安静对照组比较,三个蔬菜汁组血清GSH含量均显著增加,藕汁组GSH-Px活性显著增加,油菜汁组SOD活性显著增加;而黄瓜汁组骨骼肌线粒体MDA含量显著减少。结论:蔬菜汁可减轻游泳运动造成的脂质过氧化,增强酶性和非酶性抗氧化防御系统功能。

Abstract:Objective To observe the effects of lotus root juice,rape juice,cucumber juice with different antioxidant activities on the oxidative stress induced by swimming in rats.Methods Fifty male mature Wistar rats were randomly divided into control,swimming,lotus root juice+swimming,grape juice+swimming and cucumber juice+swimming groups(10 rats per group).Distilled water was intragastrically applied to the control and swimming groups,and fresh root juice,grape juice and cucumber juice were respectively administrated in the remaining three groups for 4 weeks.At the end of the experiment,rats in swimming group and vegetable juice groups swam by tail-slung for 6h,and then blood,heart,liver and muscle of rats were removed.Except swimming,the treatment of rats in control group was the same as other groups.The content of malondialdehyde(MDA),vitamin C and E,and GSH,total antioxidative capacity(T-AOC),mitochondria membrane potential and fluidity,and the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and catalase(CAT) in serum and/or heart,liver,and muscle were assayed.Results As compared to the control group,the MDA content in serum,heart,liver,skeleton muscle and mitochondria of skeleton muscle increased,the content of vitamin C and E,and the activity of SOD,GSH-Px and CAT in serum,and T-AOC in serum,heart,liver and skeleton muscle decreased distinctively,and gastrocnemius muscle mitochondria membrane potential and fluidity decreased in swimming group.As compared to the swimming group,the content of MDA in serum,heart,liver and skeletal muscle mitochondria decreased significantly,the content of vitamin C,GSH,and T-AOC,and the activity of SOD,GSH-Px and CAT in serum increased distinctively,and T-AOC in heart and liver increased significantly in the three vegetable juice groups.As compared to the control group,the content of GSH increased in all vegetable juice groups,the activity of GSH-Px increased in lotus root group,the activity of SOD increased in grape juice group,and the content of MDA in skeleton mitochondria decreased in cucumber juice group.Conclusion Vegetable juices with different antioxidant activities could lesson lipid peroxidation induced by swimming,and strengthen the function of enzyme and non-enzyme antioxidant defense system.
