摘    要:目的 基于网络药理学方法挖掘巴西人参核心药效,结合动物实验验证其抗疲劳作用。方法 通过文献检索方法收集巴西人参的化学成分,运用Discovery studio软件筛选类药性成分,通过Swiss target Prediction数据库对类药性成分预测分析关键靶点,并对预测靶点进行蛋白质相互作用关系分析,采用Cytoscape 3.6.1软件中的MCODE插件筛选出PPI网络中的核心靶点,通过DAVID数据库对核心靶点进行生物信息学分析。将小鼠随机分为空白对照组、阳性对照组、巴西人参高、低剂量组,各组小鼠给药15天后进行转棒实验及负重游泳实验,检测小鼠血清乳酸、肝糖原含量,考察各组小鼠抗疲劳能力。结果 网络药理学结果提示巴西人参主要有调节甾体类激素以及物质代谢的作用;巴西人参高剂量组能够显著延长小鼠的转棒疲劳时间以及力竭游泳时间(P < 0.05或P < 0.01);巴西人参高、低剂量组均能够显著降低血清中乳酸堆积、增加肝糖原的含量(P < 0.05或P < 0.01)。结论 巴西人参具有多成分、多靶点、多途径的作用特点,其核心功效可能为抗疲劳以及增强性功能作用;动物实验结果表明巴西人参具有良好抗疲劳效果,其机制可能为促进机体内乳酸的转化,增加体内储能物质即肝糖原的储备。

Abstract:Objective To excavate the core drug effects of Pfaffia paniculata based on the method of network pharmacology, as well as to verify its anti-fatigue effect combined with animal experiments.Methods We collected the chemical components of Pfaffia paniculata through literature search, used Discovery studio software to screen its drug-like components according to the drug-like principle, and used the Swiss target Prediction database to predict the target points of Pfaffia paniculata-like drug components. We performed protein interaction relationship analysis on predicted targets, used the MECOD plug-in in Cytoscape 3.6.1 software to screen out the core targets in the PPI network, and performed bioinformatics analysis on the core targets through the DAVID database. The mice were randomly divided into blank group, positive drug group, high and low dose groups of Pfaffia paniculata. The mice in each group were given medicine for 15 days. The anti-fatigue ability of mice in each group was tested by rotating rod test and weight-bearing swimming test, and the contents of lactic acid and liver glycogen were detected.Results The results of network pharmacology indicated that Pfaffia paniculata was mainly involved in the regulation of steroids and material metabolism; Pfaffia paniculata high-dose group could significantly prolong the fatigue time of the rotating rod and the time of exhausted swimming in mice (P < 0.05 or P < 0.01), Pfaffia paniculata high and low dose groups could significantly reduce the accumulation of lactic acid in the serum and increase the content of liver glycogen (P < 0.05 or P < 0.01).Conclusion Pfaffia paniculata has the characteristics of multiple components, multiple targets, and multiple pathways. Its core efficacy may be anti-fatigue and enhancing sexual function. Pfaffia paniculata has a good anti-fatigue effect, and its anti-fatigue mechanism may be to promote lactic acid and increase the production of liver glycogen.
