摘    要:目的建立人血浆样品中盐酸二甲双胍浓度的含量测定方法。方法血浆以10%三氯乙酸作为沉淀剂.以Symmetry C18(46mm×150mm,5um)为色谱柱,乙腈:水:3mmol·L^-1磷酸盐缓冲液(含0.1%庚烷磺酸钠和0.5%三乙醇胺。pH4.5)(7:43:50)为流动相,流速为1.0mL·min^-1,柱温30℃,检测波长为233nm。结果盐酸二甲双胍浓度在1~100μg·mL^-1范围内线性关系良好,回归方程为A=9.627416×10^4C.7.9609643×10^3(r=0.9998),平均回收率为99.21%。结论本法简单、快捷、灵敏、准确,适用于临床药代动力学及药效学的研究。

Abstract:OBJECTIVE To develop a method for the determination of hydrochloride metformin in human plasma. METHODS Samples of plasma were precipitated with trichloroacetic acid. A Symmetry C18 column was used with a mobile phase of acetonitrile-water-phosphate buffer (containing 0.1% sodium beptanesulfonate, 0.5 % triethanolamine and adjusted pH to 4.5) (7: 43 : 50). The flow rate was 1.0mL· min^- 1, the temperature of the column was 30℃, the detection wavelength was at 233.0nm. RESULTS A good linearity was obtained from 1 to 100μg·mL^-1 plasma concentration of hydrochloride metformin A = 9. 627416 × 10^4C-7. 9609643 ×10^3 (r = 0. 9998). The mean recovery was 99.21%. CONCLUSION The method was simple, rapid, accurate, and suitable for being applied to study of pharmacokinetics and pharmacodynamics of metformin hydrochloride in clinical.
