摘    要:目的研究芎芍胶囊含药血清对人脐静脉内皮细胞(HUVEC)黏附分子表达的影响。方法采用血清药理学方法,制备芎芍胶囊含药血清体干预体外培养的HUVEC,MTT实验检测其对HUVEC增殖的影响,采用RT-PCR和ELISA法测定HUVEC黏附分子VCAM-1、ICAM-1、ICAM-3、E选择素的表达水平。结果芎芍胶囊含药血清的低、中、高剂量组细胞的OD值明显高于空白血清组,同时含药血清组VCAM-1、ICAM-1、E选择素的表达和分泌水平也显著提高,而ICAM-3则无明显变化。结论芎芍胶囊对黏附分子(VCAM-1、ICAM-1、E选择素)的表达有调节作用,可能是其促血管新生的重要作用机制之一。

Abstract:Objective To study the impacts of xiongshao capsule contained serum on the expression of cell adhesion molecule(CAM)in human umbilical vein endothelial cell(HUVEC).Methods The serological pharmacological method was applied.HUVEC were incubated in vitro by the intervention of xiongshao capsule contained serum.RT-PCR and ELISA were adopted to determine the expressions of HUVEC adhesion molecule VCAM-1,ICAM-1,ICAM-3 and E-selectin.Results Compared with the control group,OD values were obviously higher in xiongshao capsule contained serum groups of low,medium and high doses.Meanwhile,the expression of CCAM-1,ICAM-1 and E-selectin as well as the secretion level were improved in drug-contained serum groups significantly.But,there was no apparent change in ICAM-3.Conclusion Xiongshao capsule can adjust the expression of adhesion molecules(VCAM-1,ICAM-1 and E-selectin),which is probably one of the important mechanisms on the promotion of angiogenesis.
