摘    要:目的研究大鼠面神经缺血后面神经元中MDA和MnSOD含量变化。方法利用外科手术方法阻断大鼠鼓室段岩动脉导致面神经缺血,造成缺血损伤后,在损伤后的1d、3d、1w、2w、3w用酶标仪测定吸光度来测定MDA的表达;用WesternBlot法测定面神经元MnSOD蛋白表达。结果 MDA含量在缺血后开始升高,在1w达到高峰,随后开始减少,差异有统计学意义(P〈0.05);MnSOD含量在缺血后开始减少,在1w达到最低值,随后开始升高,差异有统计学意义(P〈0.05)。结论面神经缺血损伤后,面神经元MDA的表达逐渐升高,而MnSOD的表达逐渐降低,两者在7d达到极点,随后逐渐恢复。说明ROS和MnSOD综合调节神经元的活性。

Expressions of MDA and MnSOD after facial nerve ischemia in rats

Abstract:Objective To study the expressions of MDA and MnSOD after facial nerve ischemia in rats. Methods Using surgical methods blocked the rat tympanic segment rock artery which could lead to facial nerve ischemia. After the facial nerve ischemia, absorbance was measured with a microplate reader to determine the expression of MDA and Western blot determine MnSOD protein expression at the time points of 1d,3d,1w,2w,3w after the facial nerve ischemia. Results MDA content increased after ischemia and peaked at 1w, then started to decrease, difference was statistically significant (P<0.05). MnSOD content started to decrease after ischemia and peaked at 1w, then started to increase. Conclusion After the facial nerve ischemia, the expression of MDA gradually increased while the expression of MnSOD gradually decreased, both of them peak at 7d, and then gradually restored. This indicates that ROS and MnSOD comprehensively regulate the activity of the neurons.
