摘    要:目的建立人体血清中5-FU含量的测定方法。方法高效液相色谱法测定人血清中5-FU的血药浓度,色谱条件:C18(250mm×4.6mm,5μm),流动相:甲醇:水(15:85),检测波长:265nm,流速:0.8ml/min,进样量:20μl。结果5-氟尿嘧啶在1-20μg/ml的线性关系良好(r=0.9993),在1、5、20μg/ml的日内精密度分别为RSD=5.63%、RSD=2.14%、RSD=2.32%。在1、5、20μg/ml的回收率分别为76.76%、107.87%、98.96%。结论该方法适合应用于人血清中5-FU血药浓度测定。

STUDY ON APPLICATION OF HIGH PERFORMANCE LIQUID CHROMATOGRAPHY METHOD FOR DETERMINING THE CONTENT OF 5-FU IN HUMAN SERUM

Abstract:Objective To establish the method for determination the concentration of 5 -FU in human serum. Methods The concentration of 5 -FU in human serum was determinated by HPLC method, chromatographic condition : C 18 chromatographic column ( 250 mm ×4.6 mm,5 um) ; moving phase methanol : water ( 15 : 85 ) ; detection wavelength: 265nm; flow rate: 0. 8ml/min; sample size: 20 ul. Results 5 -FU in 1 -20 ug/ml has a good linear relationship( r = 0. 9993 ). In 1, 5, 20 ug/ml, the RSD of intra - day were 5.63 % ; 2. 14% and 2. 32% respectively. The average recoveries were 76. 76% , 107. 87% and 98.96% respectively. Conclusion This method is suitable for determination of the concentration of 5 - Fu in human serum.
