黄连温胆汤改善2型糖尿病大鼠糖脂代谢紊乱

摘    要:目的:考察黄连温胆汤改善2型糖尿病引起的大鼠糖脂代谢紊乱作用。方法:通过60天高糖高脂饲料喂养合并一次性35 mg·kg-1链脲佐霉素(STZ)腹腔注射制备2型糖尿病大鼠模型,测定血糖,筛选模型成功大鼠。将该大鼠分为空白组、模型组、二甲双胍组(0.2 g·kg-1)和黄连温胆汤3、6、12 g·kg-1三个剂量组,连续给药30天。测定血清中空腹血糖(FBG)、血清胰岛素(FINS)、糖化血红蛋白(GHB)、胰岛素敏感系数(ISI)、胰岛素抵抗系数(IRI)、甘油三酯(TG)、总胆固醇(CHO)、低密度脂蛋白胆固醇(LDLC)和高密度脂蛋白胆固醇(HDLC),评估黄连温胆汤对大鼠糖脂代谢紊乱的调节作用。HE染色后观察胰腺病变。结果:黄连温胆汤能降低FBG、GHB,增加ISI,降低IR I;降低血清TG、CHO和LDLC含量,升高HDLC含量;但对FIN S作用不明显。此外,黄连温胆汤与模型组相比减轻了胰腺细胞变性,改善胰岛病变。结论:黄连温胆汤可以有效地改善2型糖尿病引起的糖脂代谢紊乱,保护胰腺细胞,其作用机制可能与改善胰岛素敏感性和胰岛素抵抗有关。

Abstract:Objective: The aim of this study is to investigate the therapeutical effect of Huanglian Wendan decoction on metabolism disorder in rat model of type 2 diabetic mellitus. Methods: 60 days high-glucose-high-fat diet and 35 mg/kg streptozocin (STZ) injection once were used to bulid a rat model of type 2 diabetic mellitus.Following, metformin hydrochloride (0.2 g/kg), berberine hydrochloride (0.2 g/kg), HLWDD(3, 6, 12 g/kg) were given by gavage for 30 days except normal and model groups after randomly divided into six groups. Relate measurements about glucose and lipid metabolism in serum were did, and insulin sensitivity index (ISI) and insulin resistance index (IRI) were calculated to evaluate the improvement of Huanglian Wendan decoction on metabolism disorder. The pathological damage of pancreas was observed by a HE staining. Result: 30 days administration of Huanglian Wendan decoction to the rats reduced fast blood glucose (FBG), glycosylated hemoglobin(GHB), IRI, triglyceride (TG), total cholesterol (CHO) and low density lipoprotein cholesterol (LDLC) in serum; increased ISI and high density lipoprotein cholesterol (HDLC). However, its effect to FINS can not significantly observed. Treatment of HLWDD to rats also protected them from pathological damage of pancreas. Conclusion: Treatment of Huanglian Wendan decoction protects rats from metabolism disorder and pathological damage of pancreas in a rat model of type 2 diabetic mellitus significantly. Its mechanism may related to the improvement on insulin sensitivity and insulin resistance.
