血管内皮生长因子与育龄妇女卵巢血流和功能关系的研究

摘    要:目的:探讨育龄妇女卵巢VEGF表达与E2、P分泌、卵巢血流、血清VEGF水平的关系。方法:选择19例卵巢功能正常的子宫肌瘤育龄妇女患者,应用多普勒超声测定优势卵泡所在侧卵巢内血流阻力指数、搏动指数、收缩峰值血流速度(PSV);用 ELISA法测定血清VEGI水平;用放射免疫双抗法测定血清比P水平;用免疫组化法测定优势卵泡或黄体VEGF表达强度。结果:(1)卵巢VEGF表达在整个月经周期与血E2水平呈显著正相关(r=0.502,P<0.05),在黄体期与血清P水平呈显著正相关(r=0.837,P<0.01)。整个月经周期内血清VEGF水平与血清E2浓度无相关性(P>0.05),黄体期血清VECF水平与血清E2、P浓度呈正相关(r分别为0.565和0.625,P<0.05);(2)卵巢VEGF表达与优势卵巢内血流搏动指数、阻力指数呈负相关(r分别为-0.615和-0.646,P均<0.05);(3)育龄妇女卵巢VEGF表达强度与血清VEGF水平呈显著正相关(r=0.720,P<0.01)。结论:卵巢VEGF表达与E2、P分泌功能有关,其机理可能是通过旁分泌和自分泌方式分别作用于血管内皮和性激素分泌细胞;卵巢VEGF表达与优势卵泡侧卵巢内血流有关;育龄妇女卵巢VEGF表达与血清VEGF浓度呈显著正相关,卵巢是影响育龄妇女血清VEGF水平的主要器官之一。

Abstract:ve: To study the relationships between expression of vascular endothelial growth factor (VEGF) in the ovary and serum VEGF level,ovary blood flow,ovary function.Meth-ods: 19 reproductive-aged women were studied. Ovary blood flow including pulse index (PI), resis-tance index ( RI), peak systolic velocity (PSV) was measured by color doppler sonography. Serum levels of E2, P were measured by radioimmunoassay and serum VEGF level by ELISA. Expression of VEGF was determined by ABC immunohistochemistry assay. Results: To normal reproductive-aged women, intensitiy of VEGF expression in ovary was correlated with estradiol concentration in whole menstrual period ( r = 0. 502, P < 0. 05 ) and significantly correlated with progesterone level in luteinic phase ( r = 0.838, P < 0.01) . There was also positive correlation between serum VEGF lev-els and serum concentrations of estradiol ( r = 0.565, P < 0.05) as well as progesterone in luteinic period ( r = 0.625, P < 0. 05). Intensity of VEGF expression in ovary was significantly correlated with serum VEGF level (r = 0.720,P<0.01) and negatively correlated with PI and RI ( r = - 0.615 and - 0.646 respectively, P < 0.05). Conclusions: VEGF in the ovary is correlated with hormone-secreting and blood flow of dominant ovary. Expression of VEGF in ovary is significantlycorrelated with serum VEGF levels and ovaries are one of the main organs determining serum VEGF concentration of reproductive-aged women.
