丹皮酚对乳腺癌细胞的生长抑制作用

摘    要:[目的]研究丹皮酚(Pae)对人乳腺癌细胞的作用及机制。[方法]采用MTT法检测Pae对乳腺癌Bcap37、MDA-MB.453细胞的增殖抑制作用,流式细胞仪检测Pae对Bcap37和MDA-MB-453细胞凋亡和周期分布的影响,WesternBlot检测相关蛋白表达。[结果]Pae对乳腺癌Bcap37及MDA-MB-453细胞均具有增殖抑制作用,并呈时间和剂量依赖性;流式细胞仪分析细胞凋亡率呈浓度依赖性,细胞周期分析显示Pae阻滞Bcap37及MDA-MB-453细胞在G1期;WestemBlot检测凋亡相关蛋白Caspase7、Caspase3和PARP表达减少,而P-FEN蛋白表达增加。[结论]Pae对乳腺癌细胞系具有增殖抑制作用,其机制可能与促进细胞凋亡及引起细胞周期阻滞。PrEN表达上调有关。

Effects of Paeonol on Growth Inhibition of Human Breast Cancer Cells

Abstract:[Purpose] To investigate the effect of Pae on proliferation of human breast cancer cells and its possible influencing mechanism. [Methods] MTr assay was used to measure the effect of Pae on proliferation of breast cancer cells Bcap37 and MDA-MB-453. Apoptosis and cell cycle were measured by flow cytometry. Western Blot was used for detection of protein ex- pression. [Results] Pae showed inhibitory effect on Bcap37 and MDA-MB-453 breast cancer cells in time- and dose-depender~t.Bcap-37 and MDA-MB-453 cells treated by Pae were blocked in G~ phase. The apoptosis rate appeared in dose-dependent.Western Blot showed that the expressions of Caspase 3, Caspase 7 and PARP decreased and the expression of PTEN in- creased. [Conclusion] Pae can inhibit the growth of breast cancer cells,which is possibly con- tribute to apoptosis inducing and cell cycle arrest.
