抑癌基因PTEN在胃癌中的表达及其生物学意义

目的　探讨抑癌基因PTEN蛋白在胃癌组织中的表达状况及其生物学意义。方法　采用免疫组化染色方法检测 6例正常胃黏膜 ,2 4例早期胃癌及 3 0例中晚期胃癌组织中PTEN蛋白的表达。结果　正常胃黏膜组织中PTEN蛋白高表达( 10 0 % ) ,胃癌组织中PTEN低表达 ,且阳性强度较低 ,两者差异显著 (P <0 0 5 )。胃癌组织中PTEN蛋白表达与肿瘤分期及分化程度无明显关系 ,但有淋巴结转移者PTEN蛋白阳性率仅为 9 5 % ,而无淋巴结转移者PTEN蛋白表达阳性率达 69 7% ,两者间差异显著 (P <0 0 1)。结论　胃癌组织中存在着抑癌基因PTEN蛋白表达缺乏或异常 ,PTEN蛋白表达可能与胃癌的发生及生物学行为有关 ,抑癌基因PTEN是胃癌的一种新的相关基因

Expression and significance of tumor suppressor gene PTEN in gastric carcinoma

Objectives To investigate expression and significance of tumor suppressor gene PTEN in gastric carcinoma. Method Immunohistochemical method was used to detect the expression of PTEN gene in 54 cases of gastric carcinomas and 6 cases of normal gastric mucous membrance tissues.Results The positive rate of PTEN protein was 100% in normal gastric tissues, but 46.3% (25/54) in gastric carcinomas. most of them were weak positive.The results showed a statistical significance between normal gastric tissues and gastric carcinomas(p<0.05). The positive rates of PTEN protein which were positive or negative lymph node metastasis of gastric carcinoma was 9.5%(2/21) and 69.7%(23/33) respectively.The difference was significant(p<0.01).Conclusion Expression of PTEN protein in gastric carcinoma was different from that of normal Gastric tissues. Some gastric carcinoma tissues had weak or negative expression of PTEN protein. This study suggest that there was inactivation of tumor suppressor gene PTEN in gastric carcinoma and the inactivation of tumor suppressor gene PTEN is related to tumorigenesis of gastric carcinoma. Tumor suppressor gene PTEN is probably a new tumor suppressor gene which related to gastric carcinoma.
