天花粉蛋白诱导人宫颈癌HeLa细胞调亡的分子机制研究

摘    要:目的:利用基因芯片技术研究天花粉蛋白对人宫颈癌Hela细胞基因表达谱的影响,以深入探讨其作用的分子机制.方法:提取药物处理前后HeLa细胞RNA并逆转录成cDNA,在获得的cDNA上分别标记CY3和CY5两种荧光物质,然后与BiostarH-I 表达谱芯片杂交,扫描后对获得的数据用GenePix Pro 3.0软件分析.结果:两组间存在差异性表达基因78条,其中与细胞黏附及细胞间相互作用相关的MGST3、LGALS3BP等16条基因表达下降,与细胞凋亡密切相关的NOP56、TNFSF10、CASP9、DFFB等62条基因表达上调.结论:基因芯片技术能高通量分析天花粉蛋白作用于HeLa细胞的相关靶基因,为进一步揭示天花粉蛋白的抗癌机制提供了新的思路.

Abstract:Objective:To detect the gene expression profiles of human cervical carcinoma Hela cell with microarray technique after treated by Trichosanthin and to further investigate its molecular mechanism .Methods:RNA from both control and treated cells were isolated and reversely transcribed to cDNA with the incorporation of cy3 and cy5-labeled dUTP, the probes were then hybridized with BiostarH-I cDNA microarray, the chip was scanned and then analyzed by GenePix Pro 3.0 software.Results:there were 78 pieces of genes changed distinctly, 16 genes associated with adhesion and interaction between cells were down-regulated and 62 genes closely related with apoptosis were up-regulated.Conclusion:DNA microarray can find the target genes of the TCS's effect on Hela cells and can put light on its anti-cancer mechanism study.
