| 摘 要： | 一、Hedgehog通路概况
Hedgehog (Hh)信号通路最初是在果蝇中发现的,该通路非常保守,在胚胎发育过程中,其决定着许多关键细胞的命运如增值、分化、凋亡、迁移等,从而广泛调节胚胎形成、发育以及众多组织的重塑1-2].在成年时的许多组织,包括肝脏的损伤修复中发挥着重要作用3-6].该信号通路主要包括Hh生成细胞和Hh反应细胞,前者能产生一个配体家族包括Sonic hedgehog (Shh),Indian hedgehog (Ihh),Desert hedgehog (Dhh);然后通过自分泌、旁分泌和内分泌途径作用于后者,并与其细胞膜上的受体Patched (Ptc)结合,同时通过抑制Smoothened (Smo)分子活性启动胞内信号通路,导致Glioblastoma (Gli)家族转录因子(Glil,Gli2,Gli3)前往细胞核内,调节Gli靶基因的表达5-7].
