摘    要:目的:探索食物纤维成分之一丁酸钠对人大肠癌细胞生长,分化的影响。方法:应用MTT、流式细胞仪、光镜及电镜技术、观察经丁酸钠诱导后低分化大肠癌细胞系Clone-A细胞生长及形态学变化。结果:经丁酸钠诱导后,Clone-A细胞生长受到明显抑制:G0/G1期细胞百分比增加,S期细胞百分比减少,细胞增殖指数下降,诱导后Clone-A细胞变长,出现胞浆突起,由圆形、不规则形变为扁平的成纤维细胞样形态,同时,丁酸钠作用2d后,细胞表面伸出丝状伪足,4d后明显变长,胞浆内质网、高尔基体数量及密度增加,内质网集核周围并铺展于胞质内,形态趋于正常。结论:丁酸钠能够抑制低分化人大肠癌细胞生长,诱导其成熟分化,使其一些恶性表型逆转。

Abstract:Objective:This paper was to study the effect of production butyrate in dietary fiber on the proliferation and differentiation of human colorectal cancer cells. Methods:We used MTT assay, flowcytometry, light microscopy, scanning and transmission electronic microscopy to observe the growth and morphological changes in poorly differentiated human colorectal cancer cell line, Clone A,treated by sodium butyrate. Results:Sodium butyrate significantly inhibited the growth of Clone A cells and caused a G 0/G 1 cell cycle arrest. The cell formed long processes and altered from round and irregular shape to flatten fibroblast like appearance. The Clone A cells grew more pseudopodia on the surface 2 days after the treatment and exhibited more long pseudopodia 4 days after treatment. The endoplasmic reticulum and Golgi apparatus increased in number and density, with endoplasmic reticulum clustering in perinuclear area and extending in cytoplasm in normal state. Conclusion:Sodium butyrate may induce differentiation and reverse some malignant phenotype of poorly differentiated human colorectal carcinoma cell.
