熄风定颤汤对PD模型大鼠单氨类神经递质的影响

摘    要:目的:观察熄风定颤汤(以下简称中药)对PD模型大鼠旋转行为和纹状体单氨类神经递质的影响,探讨其可能作用机制。方法:采用6-OHDA偏侧纹状体两点注入法制备PD模型大鼠,随机分为模型组、美多巴组、中药高、中、低剂量组,并设假手术组为对照组,灌胃治疗4周。观察各组大鼠治疗前后APO诱导的行为学变化及对纹状体单胺类神经递质的影响。结果:美多巴组灌胃后2周旋转行为有所下降(P<0.01),但4周后又有所增加(P>0.05);中药高、低剂量组灌胃后4周后旋转行为有明显改善(P<0.05,P<0.05);中药中剂量组灌胃后2周、4周后旋转行为均有明显改善(P<0.05,P<0.01)。美多巴组、中药高、中、低剂量组大鼠损毁侧纹状体DA含量、HVA、DOPAC含量均明显增加,其中中剂量组增加最明显,分别比损毁对侧下降了21.78%、23.53%、3.52%,与损毁对侧、假手术组损毁侧差异不显著(P均>0.05),与模型组大鼠DA和DOPAC相比差异显著(P<0.01,P<0.05)。结论:熄风定颤汤可明显改善PD模型大鼠的旋转行为、增加纹状体内DA及其代谢产物的含量,以中剂量组效果最明显。

Research on the Effects of Xifengdingchan Tang on Monoaminergic Neurotransmitter in PD Model Rats

Abstract:Objective:To observe the effect of Xifengdingchan tang on rotational behavior,striatal monoaminergic neurotransmitter of PD rats and analyze its effective mechanism. Methods :PD model rats were made by injecting 6 -OHDA twopoint into unilateral striatum, randomly divided into model group, Madopar group, herbal medicine high, medium and low dose group, sham - operated group established for the control group ,treatment lasted for 4 weeks. The ethological change of PD model rats induced by apomorphine (APO) and stfiatal monoaminergic neurotransmitter were observed before treatment and 4 weeks' treatment. Results: The rotational behavior of PD in Madopar group decreased after 2 weeks' treatment ( P 〈 0.01 ) , but increased after 4 weeks' treatment ( P 〉 0.05 ) ; in high and low dose group improved obviously after 4 weeks' treatment ( P 〈 0. 05, P 〈 0. 05 ) ; in medium dose group improved obviously both after 2 and 4 weeks' treatment ( P 〈 0. 05, P 〈 0. 01 ). The content of dopamine (DA) , homovanillic acid(HVA) ,dihydroxyphenylacetie acid(DOPAC) in damaged lateral were all markedly increased in Madopar group, high,medium and low dose group;the medium dose group increased the most obviously, respectively decreased 21.78% ,23. 53% ,3.52% compared with the contralateral;there was no significant difference compared with the eontralateral and sham group ( P 〉 0.05 ) ; difference of DA and DOPAC were significant compared with model group, ( P 〈 0. 01, P 〈 0. 05 ). Conclusion : Xifengdingchan tang could significantly improve the rotational behavior in PD rats and increase the content of striatal DA and its metabolites,medium dose group was most obviously.
