含抗幽门螺杆菌特异性抗体免疫牛奶的制备

目的 :寻求幽门螺杆菌 (Helicobacterpylori,Hp)感染的防治方法。方法 :应用空肠弯曲菌选择培养基对胃镜取出的胃粘膜组织进行Hp培养及细菌学检查 ,用该菌免疫牛 ,采用 0、15、30、4 5天四次接种免疫 ,制备含高效价的抗Hp抗体的牛奶。 结果 :在牛被免疫 4 5天时 ,经琼脂免疫双扩散法检测 ,牛血中抗Hp抗体效价为 1:32 ,牛奶中抗Hp抗体的效价为 1:16。牛被免疫 5 0天时 ,琼脂免疫双扩散法检测 ,牛血和牛奶中抗Hp抗体效价均可达 1:32 ;酶联免疫试验 (ELISA)法检测 ,牛被免疫 5 0天时 ,牛血中抗Hp抗体效价可达 1:5 12 0 0 ,牛奶中抗Hp抗体的效价为 1:10 2 4。结　论 :经幽门螺杆菌免疫后的奶牛 ,在免疫 4 5天后即可获得高效价的抗Hp抗体牛奶。该方法为Hp感染的特异性防治开辟了美好的前景。

Preparation of Immune milk containing Anti-Helicobacter pylori Specific Antibody

Objective: Scarch for specific preventing and ther ap y method of Hp infection. Methods Helicobacter pylori (Hp) obtained from the gas tric mucous menbrane tissue using gastrosopy operation was cultivated in the sel ective medium for Campylobacter jejuniand. Examining Hp using morphologic diagno stic method. Obtaining the specific milk of high titer antibody against Hp after immunizing the cow with the Hp four times (0,15,30,45 day). Results: Using the double gel diffusion test, the titer of antibody in cow blood i s 1:32,while in milk is 1:16 after 45 day immunizing. However the titers of anti body in cow blood and milk are both 1:32 after 50 day immunizing. Comclu sion: The specific milk of containing high titer antibody against Hp ca n be obtained after 45-day immunizing cow using Hp. This method opens up a prosp erous way to the specific prevention and therapy of Hp infection.
