前列地尔调节Bc1-2,Bax抑制大鼠糖尿病肾病肾小管细胞凋亡

摘    要:目的 探讨前列地尔对大鼠糖尿病肾病(DN)肾小管细胞凋亡及蛋白Bc1-2/Bax表达的影响.方法 雄性Wistar大鼠30只随机抽取10只作为正常对照组,余20只制备成糖尿病大鼠模型,并将成模大鼠随机分为糖尿病组和前列地尔组.16周后取大鼠肾脏皮质,免疫组化法检测肾脏组织Bc1-2/bax蛋白表达,TUNEL检测肾脏细胞的凋亡.结果 与正常对照组相比,前列地尔组和糖尿病组Bc1-2和bax表达水平均升高,Bc1-2/bax比值降低,肾小管细胞凋亡指数增加(P<0.05).与糖尿病组比较,前列地尔组Bc1-2表达量及Bc1-2/bax比值升高,bax蛋白含量和肾小管细胞凋亡指数降低(P<0.05).结论 前列地尔可通过调节DN肾脏组织中Bc12,bax的表达,发挥对肾脏的保护作用.

The Effects of Alprostadil on Apoptosis of Kidney Tubules Cells Through Regulation of Bcl-2/Bax in Diabetes Mellitus Rats

Abstract:Objective To explore the effect of alprostadil on apoptosis of kidney tubules cells and expression of bcl-2/bax protein in Diabetes Mellitns Rats. Methods Thirty healthy male Wistar rats were enrolled,ill which 10 were randomly assigned to normal control group, and the others were established into diabetes mellitus( DM ) model. The twenty rats model were assigned to DM model group and alprostadil group. After sixteen weeks,rat kidneys were taken and the change of bcl-2/bax was detected by immunohistochemistry, as well as apoptosis of kidney tubules cells by terminal-deoxynucleotidyl transferase mediated dUTP nick end labeling( TUNEL). Results The levels of hcl-2, bax protein and kidney tubules cells apoptosis index were higher,while bcl-2/hax ratio was lower in alprostadil and DM group than those in normal control group ( P 〈 0.05 ). The levels of bcl-2 protein, bcl-2/bax ratio and kidney tubules cells apoptosis index were higher in alprostadil group than those in DM group ( P 〈 0.05 ) , and bax expression level was reduced compared to that of DM group( P 〈 0.05 ). Conclusion Alprostadil can protect Diabetes Mellitus Rats' kidney through regulating the expression of bcl-2/bax protein.
