三氧化二砷抑制裸鼠胃癌腹腔种植腹水生成

 目的　研究As2 O3对裸鼠胃癌腹腔种植腹水生成的影响及其作用机制。方法　 10 0只BALB/C nu/nu裸鼠腹腔内接种胃癌MKN 4 5细胞株后 ,随机分为 5组 ,然后分别腹腔内注射生理盐水、表阿霉素及不同剂量的As2 O3,观察癌细胞株对各组裸鼠的致瘤率、腹水生成量及生存时间的影响 ;透射电镜观察As2O3作用后腹腔内肿瘤细胞的形态学变化。结果　表阿霉素、低剂量As2 O3与生理盐水对照组相比可显著抑制裸鼠腹水的生成 ,延长生存期 (P <0 .0 1) ;中高剂量As2 O3还可通过诱导肿瘤细胞凋亡消除腹腔内肿瘤细胞、抑制腹水产生、而使裸鼠寿命明显延长。结论　As2 O3可通过诱导胃癌细胞凋亡而抑制或消除裸鼠胃癌腹腔种植及腹水的生成 ,不同程度地延长生存期。

The inhibition effection of As2O3 for nude mice ascitis production planted by gastirc cancer cell line

Objective To observe the effects of As 2 O 3 on malignant\|ascites of nude mice and probe into its mechanism of action.Methods BALB/c\|nu/nu nude mice were injected with MKN\|45 gastric cancer cell line and were divided into 5 groups at random : normal saline , Epirubicin and various concentration groups of As 2 O 3 . The rate of tumor formation, production of ascites and survival period were evaluated. Morphorlogical changes of tumor cell were studied by electromicroscope.Results Epirubicin and low dose of As 2 O 3 can obviously inhibit the production of ascites and prolongs the survival period of tumor\|loading nude mice ( P <0.01). Mid to high doses of As 2 O 3 can erate cancer cells through the way of apoptosis.Conclusion As 2 O 3 can induce gastric cancer cell apoptosis, inhibit or erate production of ascites and prolong the survival period of nude mice that suffered from malignant ascites from metasasis.
