摘    要:目的 探讨端粒长度与高等真核细胞生物中正常体细胞增殖能力间的关系。方法 以不同年龄的正常腮腺组织及正常皮肤表皮组织做标本,利用Southern杂交方法测定每份标本的端粒限制性片段长度,然后比较同一组织不同年龄的端粒限制性片段长度,并进行统计学分析。结果 同一年龄不同组织的端粒酶限制性片段长度是不同的,皮肤表皮的端粒限制片段长度平均值随年龄增长而变短;腮腺组织的端粒限制性片段长度平均值未随年龄增长而变短。结论 端粒缩短是细胞复制性衰老的主要原因,在不同的组织中,由端粒缩短引起的细胞复制性衰老表现程度是不同的。

Abstract:Objective To investigate the correlation between telomere restrict fragment length and proliferative ability of normal somatic cell in higher eukaryote and the important role that telomere reduction played in cell senescence.Methods The samples were collecled from normal parotid tissue and epidermis of skin from different-aged donors.The telomeric restrict fragment (TRF) lengths of the simples were measured by southern blot.The TRF length of the same tissue from different-aged donors were compared and analysed statistically.Results The TRF length in different tissues from the same-aged donors were not accordant.The mean value of TRF length in epidermis of skin became short with the age of donors increasing;but the mean TRF value in parotid didn't.Conclusion Telomere reduction is an important factor during cell senescence.In different tissues,the cell replicative senescence degree caused by telomere reduction is various.
