摘    要:目的:观察骨髓基质细胞(bone marrow stromal cells,BMSCs)移植对大鼠脊髓损伤后环氧化酶-2(cyclooxygenase-2, COX-2)和血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白表达的影响。方法大鼠随机分为3组:A为假手术组,B为细胞培养基(DMEM)对照组,C为BMSCs移植组。B组和C组大鼠进行脊髓压迫损伤模型制作。脊髓损伤后, B组和C组大鼠分别给予DMEM培养液或BMSCs脊髓损伤周围区注射。术后应用Western Blot方法检测COX-2、VEGF蛋白在损伤脊髓组织中的表达变化。结果 COX-2、VEGF蛋白在A组大鼠未损伤脊髓组织中均低水平表达。B组与A组相比,脊髓损伤后COX-2蛋白表达水平明显增加(P<0.01),VEGF蛋白表达水平则短暂显著增加(P<0.05)。C组与B组相比, COX-2和VEGF蛋白表达水平均显著增加。结论 BMSCs移植能够增加脊髓损伤后COX-2、VEGF蛋白的表达。

Abstract:Objective To observe the effect of bone marrow stromal cells (BMSCs) transplantation on expression of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) protein in rats following spinal cord injury (SCI). Methods Adult rats were randomly divided into group A (sham operation group), group B (DMEM control group), and group C (BMSCs transplantation group). A rat spinal cord compression injury model was established. Rats in groups B and C were injected with BMSCs or DMEM after SCI. Expression of COX-2 and VEGF protein in rats following SCI was detected by Western blot. Results The expression level of COX-2 and VEGF protein was low in group A. The expression level of COX-2 and VEGF protein was significantly higher in group B than in group A and in group C than in group B (P〈0.01, P〈0.05). Conclusion BMSCs transplantation can up-regulate the expression of COX-2 and VEGF protein in rats following SCI.
