脐带血血清体外培养的人骨髓间充质干细胞对干细胞移植病人造血重建的影响

目的：观察脐带血血清体外培养体系培养的人骨髓间充质干细胞对干细胞移植病人造血重建的影响及不良反应。方法：用10%脐带血血清体外培养体系培养的人骨髓间充质干细胞联合自体外周血干细胞移植治疗3例恶性淋巴瘤患者,计数BM-MSC的数目,观察其造血重建时间、不良反应。结果：采用10%脐带血血清培养体系培养BM-MSC,回输时达到2.82-3.96×106/kg,3例患者中性粒细胞〉0.1×109/L的时间为移植后第9-12天,血小板〉20×109/L的时间为移植后第9-15天,无明显不良反应。结论：10%脐带血血清培养体系是一种基于临床移植需要的分离、培养人BM-MSC的方法,此法培养的BM-MSC联合APBSC移植治疗恶性淋巴瘤患者,加速了造血重建,未见明显不良反应,具有临床应用价值。

Effect of bone marrow mesenchymal stem cells cultured by umbilical cord blood serum in vitro in hematopoietic recovery after autologous peripheral blood stem cells transplantation

Objective：To explore the efficacy and safety of bone marrow mesenchymal stem cells（BMSC） cultured by umbilical cord blood serum in vitro in hematopoietic recovery for the patients received autologous peripheral blood stem cells transplantation.Methods： To use autologous bone marrow mesenchymal stem cells（BM-MSC）cultured in vitro by a culture system which medium contains 10% umbilical cord blood serum in combination with peripheral blood stem cells（PBSC）transplantation treat 3 patients diagnosed with lymphoma,take count of the BM-MSC,to observe the hematopoietic recovery time and side-effect.Results： Cultured BM-MSC with umbilical cord blood serum,the count of BM-MSC ranged from 2.82×106/kg to 3.96×106/kg when reinfused into body,3 patients＇ hematopoietic recovery time was 9-15 days for neutrophilicgranulocyte 0.1×109/L and 9-15 days for platelet 20×109/L after transplantation,without obvious side effect.Conclusion： The culture system which medium contains 10% umbilical cord blood serum is a method of culturing and expanding BM-MSCs and provide experimental basis for clinical transplantation,BM-MSC cultured by umbilical cord blood serum in vitro in combination with APBSC transplantation for treatment of non-Hodgkin lymphoma is safe and feasible.Hemopoietic reconstitution is rapid.
