TC-1与VEGF在非小细胞肺癌肿瘤组织和淋巴结组织中表达及相关性研究

摘    要:目的:检测TC-1蛋白和VEGF蛋白在NSCLC中的表达,分析二者表达的相关性及其与Wnt/β-Catenin通路的调控的相关性,为进一步研究TC-1基因在NSCLC中的作用及其机制提供依据。方法:采用免疫组化EnVinsion方法检测TC-1蛋白和VEGF蛋白在95例NSCLC中的表达,统计学分析表达差异性和蛋白间的相关性。结果:免疫组化结果显示TC-1阳性表达主要位于胞浆中和部分胞核,VEGF阳性表达主要位于胞浆中。TC-1、VEGF蛋白在鳞癌、腺癌中高表达,与相应正常组织表达相比差异显著(P﹤0.01)。TC-1、VEGF在NSCLC(鳞癌、腺癌)TNM不同分期的表达强度经检验差异性显著(P=0.014;P=0.011)。有淋巴结转移原发灶TC-1、VEGF蛋白的阳性率为90.57%(48/53)、92.45%(49/53),无淋巴结转移原发灶阳性率为38.09%(16∕42)、28.57%(12/42),经spearman秩相关检验差异性非常显著(P=0.000;P=0.016)。TC-1、VEGF在NSCLC中的表达率和阳性强度存在相关性(P=0.000),相关系数rs=0.691。结论:TC-1在非小细胞肺癌(NSCLC)组织中高表达,TC-1表达高低与NSCLC总的分期、淋巴结转移与否相关;TC-1与VEGF蛋白在NSCLC及NSCLC与淋巴结转移原发灶中的表达存在很好的相关性,而VEGF基因是Wnt/β-Catenin通路的靶基因,提示TC-1蛋白可能与Wnt/β-Catenin通路的调控存在相关性。

Expressions and correlation of TC -1 and VEGF in NSCLC and lymph node tissues

Abstract:Objective: TC - 1 gene is closely related to staging, differentiation, and lymph node metastasis in non - small cell lung cancer (NSCLC) , but the activation pathway and the control mechanisms of TC - 1 protein are not yet clear. Our aim of this study was to investigate the expressions of TC - 1, VEGF proteins in NSCLC, and analyze the relationship between the two genes expression and the correlation with Wnt/β - Catenin pathway. Methods: The expression of TC - 1, VEGF proteins in 95 cases with NSCLC was deteced by immunohistochemical stain. The clinical correlations between two proteins were analyzed. Results: The TC - 1 positive expression was mainly located in the cytoplasm and minimal nucleus, and the VEGF positive expression was mainly located in the cytoplasm. TC - 1, VEGF proteins were high expressed in NSCLC and there were extremely differences comparing the expression of TC - 1 ,VEGF proteins in squamous cell lung cancer, lung adenocarcinoma with those in corresponding normal tissue respectively ( P 〈 0.01 ). There were significant differences for TC - 1, VEGF expression among TNM stages in squamous cell lung cancers, lung adenocarcinoma (P = 0. 014 ; P = 0. 011 ). The positive expression rates of TC - 1, VEGF proteins in the cases with metastasis of lymph node were 90.57% (45/53), 92.45% (49/53) , however, the positive expression rates of them in the cases without were 38.09% (16/42) ,28.57% (12/42). There was signifi- cant difference between the cases with or without metastasis of lymph node by spearman test(P = 0.000;P = 0.016). There was correlation between positive and intensity of TC - 1 and VEGF expression in NSCLC (P = 0.000), while the correlation coefficients were 0. 691. Conclusion: The TC - 1 protein is overexpressed in the NSCEC, and the ex- pression rate is related with TNM stage of NSCLC and lymph node metastasis. The positive expression rate and intensity of TC - 1 protein correlates with the expression of VEGF protein in NSCLC and the cases with metastasis of lymph node. VEGF gene is a target gene of Wnt/β - Catenin pathway, which suggest that the expression of TC - 1 is closely correlated with Wnt/β -Catenin pathway in NSCLC.
