RhG-CSF对糖尿病及非糖尿病大鼠缺血性脑卒中预后及神经生长因子表达的影响

摘    要:目的探讨重组人粒细胞集落刺激因子(rhG-CSF)对缺血大鼠神经生长因子蛋白表达的影响,并观察糖尿病对其影响。方法应用线栓法制作糖尿病及非糖尿病大鼠大脑中动脉栓塞模型后,将大鼠随机分为两组,治疗组连续sc rhG-CSF(50μg·kg-1·d-1),对照组注射等量生理盐水,进行神经功能缺损评分;计数神经细胞凋亡数量,检测NGF蛋白的表达。结果治疗组大鼠的神经功能缺损评分及缺血周边区域的TUNEL阳性细胞数均低于对照组的(P<0.01),而神经生长因子蛋白免疫阳性细胞多于对照组的(P<0.01);非糖尿病组较糖尿病组更明显。结论 rhG-CSF可增加缺血性脑卒中后周边区域的神经生长因子蛋白的表达,同时抑制神经细胞凋亡,最终改善神经功能预后;糖尿病可加重脑梗死。

Abstract:OBJECTIVE To investigate the influence of rhG-CSF on the expression of NGF after cerebral ischemia rats and the effects of diabetes mellitus.METHODS Wistar diabetic rats and non-diabetic rats were subjected to middle cerebral artery occlusion and randomly devided into two groups.rhG-CSF group received subcutaneous injection of rhG-CSF 50 μg·kg~-1·d~-1 after cerebral ischemia.Neurological severity scores TUNEL staining and immunohistological assessments were performed to examine the effects of this treatment.RESULTS The neurological score and cell apoptosis significantly decreased in the rhG-CSF groups compared with the control group while the expression of NGF significantly increased,which was more obvious in non-diabetic group.CONCLUSION rhG-CSF can improve neurological function through inhibition of neuronal cell apoptosis and increasing the expression of NGF after cerebral ischemia.Diabetes can aggravate cerebral ischemia.
