摘    要:目的 探讨人类Fas基因配体 (hFasL)和Fas基因在糖尿病的发生过程中的作用。方法 利用hFasL转基因小鼠 ,用低剂量的链脲霉素 (STZ)来诱导糖尿病。应用RT PCR鉴定hFasL在转基因小鼠的胰岛中的表达。利用血糖的测定和免疫组织化学染色的观察来认定糖尿病的病变程度。结果 hFasL转基因小鼠在胰岛组织中可以表达FasL ,通过血糖的测定和胰岛组织免疫组化染色 ,证实它们相对于普通的同品系的小鼠更容易被低剂量的STZ诱导而患病。结论 Fas和FasL在糖尿病的发生过程中起着很重要的作用。

Abstract:Objective: To investigate the role of Fas-FasL pathway in the pathogenesis of streptozotocin (STZ)-induced type I diabetes mellitus. Methods: Low dose injections of STZ were used to induce type I diabetes mellitus in the CMV-human Fas-ligand (hFasL) transgenic mice. Expression of hFasL was detected by RT-PCR. Blood glucose concentration was measured with glucotrand plus blood glucose test strips. The severity of insulitis was determined by histological examination. Results: hFasL was expressed in the islets of CMV-hFasL transgenic mice. The transgenic mice (TG) were sensitive to diabetic induction than the control wild type (WT) mice by both the evaluating the concentration of blood glucose and the development of insulitis ( P <0.05). Conclusion: Fas-FasL interaction plays a significant role in the pathogenic mechanism of type I diabetes mellitus.
