丹参酮对RBL-2H3肥大细胞增殖凋亡以及活化脱颗粒的影响

摘    要:研究丹参酮对肥大细胞增殖凋亡以及活化脱颗粒的影响,深入探讨丹参酮的抗炎药理机制。应用细胞培养、流式细胞术等方法,观察丹参酮等药物对体外培养的RBL-2H3肥大细胞增殖凋亡以及活化脱颗粒的影响。结果:丹参酮I、IIA对肥大细胞增殖有明显的抑制作用,其IC50约为20μmol/L,并能明显促进细胞凋亡,且对RBL-2H3细胞活化脱颗粒有较强的抑制作用。结论:丹参酮对肥大细胞功能具有明显的抑制作用,可能是其发挥抗炎作用的一个重要方面。

Abstract:To explore the mechanism of anti-inflammatory effect of Tansicone,a monomer component extracted from Chinese medicine Salvia miltiorrhiza,the influence of Tansinone on the proliferation,apoptosis and degranulation of mast cell line RBL-2H3 was investigated by means of cell cultures and flow cytometry.It was found that Tansinone I and II showed inhibitory effect definitely on the proliferation of the mast cell line RBL-2H3 with a IC50 of about 20 μmol/L and they could obviously promote the occurrence of apoptosis in this cell line.In addition,they also exhhibited a strong inhibitory effect on the antigen-induced degranulation of mast cells.From these observations it suggests that the anti-inflammatory effect of Tanshinone may be partially due to the down-regulation mechanism in mast cells.
