柱层析法分离与纯化心磷脂

目的 建立提取、分离和纯化心磷脂的研究方法。方法 将新鲜猪心制作为肌匀浆，从中提出总脂类，分离出丙酮沉淀物，制备为酸性磷酸脂钙盐，上样于硅胶Ｇ层析注，用洗脱液进行洗脱，分部收集洗脱液，作薄层层析，合并Ｒｆ值为０．８３的洗脱液，减压浓缩即为纯化的心磷脂。结果 薄层层析鉴定心磷脂显色呈单一斑点，其Ｒｆ值与参照物的值一致，每１００ｇ新鲜猪心可获４．５０ｍｇ纯化的心磷脂。结论 本研究是一种简便、有效的分离

Separation and purification of cardiolipin from pig heart by chrotamography method.

Objective A method of established for extraction, isolation and purification of cardiolipin. Methods A muscular homogenate was prepared from fresh pig hearts.Total lipids were extracted from it.The Ca2+ salt of acidic phospholipids was prepared after the precipitation was separated with cold acetone. The Ca + salt was put on the column chromatography of silical gel. Which was eluted with elution. A thin layer chromatography of the elution was made through fraction collection of it. Then all elution of Rf value 0.83 was puttogether. As a result, purified cardiolipin was formed by means of concentration with reducing presure. Result The identification of cardiolipin showed a single sport on the layer chromatography and its Rf value was the same as the reference. The yield of purified cardiolipin is 4.50mg per lOOg fresh pig hearts tissue. Conclusion All these finders indicated that the study provides with a simple and available method with separating and purification of cardiolipin.
