以DNA测序及限制性酶切反应鉴定PCR产物特异性

目的：对PCR产物进行特异性鉴定。方法：采用DNA测序及限制性酶切反应方法。结果：DNA测序可排除PCR产物的假阳性。另一种简便易行的方法是，设计包含某一已知偏位性酶切位点的特异引物进行PCR，通过酶切反应及电泳分析鉴定其特异性。结论：此方法对PCR产物特异性鉴定十分必要。

Identify the specificity of PCR products by DNA sequencing and restriction endonuclease reactions

Objective:To identify the specificity of PCR products. Methods:DNA sequencing and restriction endonuclease reaction was used. Results: Our study indicated that pseudopositive results of PCR products were identified by DNA sequencing. Another convenient and applicable method was that PCR primers flanking a sequence with a asymmetrical restriction enzyme site was designed. Thus the PCR preducts could be identified by restriction endonuclease reaction.
